Molecular epidemiology investigation of Torque teno sus virus (TTSuVs) in Jiangsu Province

摘要 猪细环病毒(Torque tenosusvirus,TTSuVs)是近年来新发现的一种DNA病毒,广泛分布于家猪和野猪体内。为研究TTSuVs在江苏地区猪群中的分布情况,及其各分离株之间的进化关系,本实验采用PCR技术对来自江苏地区6个猪场的398份血清样品进行TTSuVs检测,挑选部分阳性样品扩增其5'-UTR基因片段,并将所获得的基因序列与国外已报道的5'-UTR基因序列进行进化树分析比较。结果显示,TTSuVs可分为TTSuV1和TTSuV2两大分支,其单一阳性检出率分别为58.4%(227/398)和48.0%(198/398),混合阳性检出率为15.4%(64/398)。TTSuV1和TTSuV2均可分为3个亚群,本实验所获得的TTSuVs分离株在各自亚群中均有分布,地域差异不明显。江苏地区TTSuVs分离株与国外某些国家的TTSuVs分离株之间有较高的亲缘关系。本实验表明,TTSuVs感染在江苏地区猪群中普遍存在,并对我国养猪产业构成潜在威胁。 Torque teno sus virus (TTSuVs) is an emerging DNA virus, widely distributed in swine and wild boar. In this study, to investigate the prevalence of TTSuVs in Jiangsu province, and the evolutionary relationship of each isolate, 398 swine sera samples from six different farms in Jiangsu province were examined by PCR method, then some positive PCR products were selected to clone the untranslated region (UTR) gene sequence, and phylogenetic tree analysis of Jiangsu TTSuVs strains and the reference TTSuVs strains in GenBank were compared. The results indicated that TTSuVs strains could be segregated into two lineages, including TTSuV1 and TTSuV2. 58.4% (227/39) and 48.0% (198/39) were positive for TTSuV1 and TTSuV2, respectively, and 15.4% (64/398) were positive for both. TTSuV1 and TTSuV2 were both classified into three subgroups, the obtaining TTSuVs strains distributed in each subgroup. In addition, some Jiangsu TTSuVs strains had higher phylogenetic relationship with the reference TTSuVs strains in GenBank. In conclusion, our study provided the evidence that TTSuVs had been widely distribuled in pigs in Jiangsu province, and it might a potential threat to swine industry in the country.
