核因子E2相关因子2在肿瘤发生机制中的作用研究进展 被引量:3

摘要 核因子-E2相关因子2(nuclear factor-E2 related factor 2,Nrf2)可通过诱导抗氧化反应元件(anti-oxidant responseelement,ARE)调控体内多种抗氧化剂、II相解毒酶和转运蛋白的表达。Nrf2下游因子参与机体氧化损伤、钙离子超载、炎症反应及细胞凋亡等反应。Nrf2的抗氧化应激作用可抵御机体外损伤,也为肿瘤防治提供了研究前景。但是近期有研究表明Nrf2及下游因子在体外多种肿瘤细胞株以及人肿瘤组织中出现过表达,对肿瘤细胞具有保护作用并促进其生长、转移。与肿瘤细胞对化疗药物的耐药性也密切相关。 The nuclear factor E2 related factor 2 (Nrf2) controls the expression of endosomatic varietal antioxidants, phase II detoxifying enzymes and transporters, through the pathway of inducing antioxidant response element (ARE). The Nrf2 downstream genes are involved in the processes of oxidative damage, calcium overload, inflammatory reaction and apoptosis, which provide a prom- ising strategy for cancer prevention. However, recent studies indicated that Nrf2 and its downstream gene products are overexpressed in many cancer cell lines and human cancer tissues. They provide cancer cells an advantage for survival, growth and transfer. Further- more, Nrf2 is thought to be responsible for acquired chemoresistance.
