摘要
目的观察参龙汤对脑缺血大鼠血清炎性因子白细胞介素-6(IL-6)、白细胞介素-8(IL-8)、细胞间黏附分子1(ICAM-1)、肿瘤坏死因子(TNF-α)的影响。方法利用双侧颈总动脉结扎的方法制备慢性脑缺血模型,50只大鼠分为:对照组、模型组、脑复康组、大剂量参龙汤组及小剂量参龙汤组。药物处理4周后,采用ELISA法检测各组大鼠血清IL-6、IL-8、ICAM-1、TNF-α含量的变化。结果脑缺血模型组大鼠的血清IL-6、IL-8、ICAM-1和TNF-α的含量较对照组显著升高。大剂量参龙汤组可显著降低血清中IL-6、IL-8、ICAM-1、TNF-α的含量,较模型组明显降低。结论参龙汤大剂量组可显著降低血清中IL-6、IL-8、ICAM-1、TNF-α的含量,减少血清中炎性因子。这可能是参龙汤对脑缺血神经损伤的保护作用机制之一。
Objective: To investigate the effect of Shenlongtang(SLT) decoction on the contents of tumor necrosisfactor-α(TNF-α) and interleukin-6(IL-6),interleukin-8(IL-8),intercellular adhesion molecule-1(ICAM-1) in blood serum of rats with cerebral ischemia model.Methods: The cerebral ischemia animal model was established by two-vessel occlusion.50 rats were randomly divided into 5 groups: control group with slam operation;model group;positive control group of pisacetam;SLT groups(low and high dosage).Four weeks later,The contents of TNF-α,IL-6,IL-8,ICAM-1 in blood serum were determined by enzyme linked immunosorbent assay(ELISA).Results: The contents of TNF-α,IL-6,IL-8,ICAM-1 in blood serum were significantly lower in the sham operation group than those in the cerebral ischemia group.The contents of TNF-α,IL-6,IL-8 and ICAM-1 in blood serum were significantly lower in the SLT high dosage group than those in the cerebral ischemia group.Conclusion: SLT has neuron protective effect by down regulation of the contents of the inflammatory cytokines in blood serum with cerebral ischemia rats.
