摘要
以核桃分离蛋白(WPI)为原料,利用Alcalase 2.4L酶解制备高活性的ACE抑制肽。以水解度和ACE抑制率为指标,通过单因素和二次回归正交旋转组合试验,优化了Alcalase 2.4L酶解核桃分离蛋白制备ACE抑制肽的工艺。得到的最佳酶解工艺条件为pH 7.94,酶解温度60℃,底物质量浓度20 g/L,酶与底物质量比3.69∶100,酶解3 h后,酶解产物的水解度达到24.78%,ACE抑制率达到76.58%,且在此条件下获得的ACE抑制肽具有一定的抗体内消化酶特性。
The preparation of angiotensin - I converting enzyme (ACE) inhibitory peptides from walnut protein isolate with Alcalase 2. 4L was investigated. The degree of hydrolysis and ACE inhibition rate were used as indicators, and the process conditions were optimized by single factor experiment and or- thogonal experiment. The optimal enzymolysis conditions were as follows : pH 7.94, enzymolysis temper- ature 60℃, substrate mass concentration 20 g/L, mass ratio of enzyme to substrate 3.69: 100, enzymoly- sis time 3 h. Under these conditions, the degree of hydrolysis was up to 24. 78%, ACE inhibition rate was up to 76. 58%, and the ACE inhibitory peptides obtained had characteristics of antidigestive enzymes.
