丹参对大鼠肺纤维化的影响研究 被引量:10

Therapeutic effect of Salvia miltiorrhiza on pulmonary fibrosis in rats

摘要 目的:观察丹参对博莱霉素(BLM)诱导肺纤维化大鼠肺内羟脯氨酸含量变化及结缔组织生长因子(CTGF)表达的影响,以探讨丹参防治肺纤维化可能的作用机制。方法:将60只雄性SD大鼠随机分为正常对照组、模型组和丹参组各20只。模型组和丹参组大鼠气管内滴注博来霉素(5mg/kg)诱导肺纤维化。造模24h后模型组大鼠用1mL生理盐水灌胃,丹参组大鼠用丹参药液10mL/kg灌胃给药。3组均正常饲养。各组分别在灌胃后第28天处死大鼠,取肺组织样本,按试剂盒方法检测肺内羟脯氨酸含量以判断肺纤维化程度;免疫组化方法检测肺组织CTGF的表达。结果:模型组大鼠肺内羟脯氨酸含量及肺组织CTGF表达均明显升高,与正常对照组比较,差异有统计学意义(P<0.01),丹参组肺组织羟脯氨酸含量及CT-GF表达均明显低于模型组,差异有统计学意义(P<0.05)。结论:丹参治疗肺纤维化的作用机制可能与其抑制结缔组织生长因子在肺组织中的表达有关。 Objective : To observe the effects of Salvia mihiorrhiza on the hydroxyproline content and connective tissue growth factor (CTGF) expression in the lung of rats with bleomycin (BLM) -induced pulmonary fibrosis and to investigate the possible mechanism by which Salvia mihiorrhiza exerts therapeutic effect on puhnonary fibrosis. Methods: Sixty male SD rats were randomly divided into normal control group (n = 20) , model group (n = 20) , and Salvia mihiorrhiza treatment group (n = 20 ). The model group and Salvia miltiorrhiza treatment group were given BLM (5 mg/kg ) by intratracheal instillation to develop a rat model of pulmonary fibrosis. Twenty- four hours later, the model group was given 1 mL saline by gavage, while the Salvia miltiorrhiza treatment group was given Salvia mihiorrhiza liquid ( 10 mL/kg). The three groups were fed conventionally, all the rats were sacrificed, and lung tissue samples were collected. The hydroxyproline content in the lung was measured by an assay kit to judge the degree of pulmonary fibrosis ; the CT- GF expression in the lung tissue was measured by immunohistochemistry. Results: The model group showod significantly increased hydroxyproline content and CTGF expression in the lung, with a significant difference compared with the normal control group (P 〈 0. 01 ). The Salvia miltiorrhiza treatment group had significantly lower hydroxyproline content and CTGF expression in the lung than the model group (P 〈 0.05 ). Conclusion: The therapeutic effect of Salvia mihiorrhiza on pulmonary fibrosis may be due to its inhibitory effect on the CTGF expression in the lung.
