摘要
目的:探讨痛风颗粒各洗脱部位中总黄酮和绿原酸的含量测定方法,为筛选该制剂有效部位和最佳提取工艺提供依据。方法:采用紫外可见分光光度法于513 nm处测定总黄酮的含量;应用高效液相色谱法测定绿原酸的含量,色谱柱:YMC-PackJ'sphere ODS-H80 C_(18)色谱柱(150 mm×4.6 mm,5μm);流动相:乙腈-1%冰醋酸梯度洗脱,流速:1.0 ml·min^(-1),检测波长:327 nm。结果:总黄酮在0.01~0.06mg·ml^(-1)浓度范围内线性关系良好。绿原酸在13~260μg·ml^(-1)范围内与峰面积线性关系良好。结论:本法简便,易行,精密度高,重复性好,能够较好的用于痛风颗粒各洗脱部位中总黄酮和绿原酸的含量测定。
Objective: To establish a method for the determination of total flavonoids and chlorogenic acid in each elution fraction of gout granules to provide the basis for screening the effective parts and the optimal extraction technology. Method: UV was used to determine the content of total flavonoids at 513 nm, and HPLC was used to determine the content of chlorogenic acid on a YMC J' sphere ODS-HS0 Cis column ( 150 mm × 4.6 mm,5 μm) with the mobile phase of acetonitrile-1% glacial acetic acid with gradient elu- tion. The flow rate was 1.0 ml min-l and the detection wavelength was 327 nm. Result: The linearity was within the range of 0.01- 0.06 mg ml- l for total flavonoids and 13-260μg ml -1 for ehlorogenic acid. Conclusion: The method is convenient, simple and reproducible, and suitable for the determination of total flavonoids and chlorogenic acid in each elution fraction of gout granules.
