摘要
目的:研究有氧运动对高脂饮食性肥胖大鼠脂肪细胞线粒体形态和动力学的影响,并对其可能的机制进行探讨。方法:将三周龄SD健康雄性大鼠随机分为正常组和高脂饮食组,后者在第8周时选取体重超出正常对照组平均体重20%的大鼠为肥胖大鼠,再分为肥胖对照组、运动干预组,第16周时处死。采用透射电镜观察脂肪细胞的线粒体形态,采用荧光定量PCR技术检测脂肪细胞中NYGGF4基因的表达水平,采用Western blot方法检测Mfn1蛋白、Mfn2蛋白、Drp1蛋白的表达水平。结果:1)肥胖组大鼠脂肪细胞线粒体体积变小、数量明显减少,线粒体嵴断裂、减少、消失,部分线粒体肿胀,甚至呈空泡状;有氧运动干预组大鼠脂肪细胞线粒体形态基本正常,与正常组接近,表现为线粒体嵴清晰可见,线粒体无明显肿胀、皱缩。2)肥胖组大鼠脂肪细胞NYGGF4mRNA、Mfn1/2蛋白表达水平均显著高于对照组,Drp1蛋白表达水平显著低于对照组;有氧运动干预组大鼠较肥胖对照组大鼠脂肪细胞NYGGF4mRNA、Mfn1/2蛋白表达水平显著降低,Drp1蛋白表达水平显著升高,差异具有非常显著性;与正常对照组相比差异无统计学意义。结论:有氧运动可使肥胖大鼠脂肪细胞线粒体的形态、分布、数量维持适宜的动态平衡,预防线粒体遭受过多损伤;有氧运动可能通过调节NYGGF4mRNA表达水平部分影响线粒体形态及动力学。初步揭示了有氧运动干预脂肪细胞线粒体形态和动力学异常在健康减脂中的作用,从新的视角为有氧运动的减肥作用提供了理论依据。
Objective : To study the effect of aerobic exercise on the adipose mitochondrial morphology and dynamics of obese rats with high fat diet and the possible mechanism. Methods: healthy male SD rats aged three weeks were randomly divided into normal group and high fat diet group. Select the ones in later group whose weight was 20% higher than the former in the 8th week. Then the rest rats were divided into obesity control group and exercise in- tervention groups, and killed at the sixteenth week. The researchers used transmission electron microscopic to ob- serve fat cell mitochondria morphology and fluorescence quantitative PCR technique to detect the expression level of NYGGF4 gene in the fat cells and Western blot method to detect the protein Mfnl/2, protein Drpl expression level. Results : 1 ) Obese rat adipocytes mitochondria became smaller in size and less in quantity. Its mitochondrial cristae witnessed fracture, reduction and disappearance. Part of the mitochondrial swelled or even became vacu- ole. The mitochondrial morphology of fat cell of rats in the aerobic exercise group was normal and approached to the level of normal group. It was reflected in the clearly visible mitochondrial cristae and no obvious swelling or shrinking. 2 ) NYGGF4mRNA and Mfnl/2 protein expression of fat cells of obese rats were significantly higher than that in the control group. The level of Drpl expression was significantly lower than the control group.NYGGF4mRNA, Mfnl/2 protein expression levels of rats in the aerobic exercise intervention group was significant- ly lower than the control group and the level of Drpl expression was significantly increased and the difference is very significant. Compared with the normal control group, the difference was not statistically significant. Conclu- sion : Aerobic exercise can enable the form, distribution and quantity of the adipocytes mitochondria morphology of obese rats maintain an appropriate dynamic balance cause obesity;Aerobic exercise can effect on mitoehondrial morphology and dynamics by regulating the expression level of NYGGF4mRNA. This study revealed the role of aerobic exercise intervention on the adipose mitochondrial morphology and dynamics abnormality, providing theo- retical basis for losing weight effect of aerobic exercise from the new angle of view.
