急性肺损伤患者外周血Th17细胞水平及功能的研究 被引量:2

摘要 目的探讨Th17细胞在急性肺损伤发病中的作用。方法以10名急性肺损伤患者和10名健康志愿者为研究对象,分别留取外周血标本。利用RT-PCR技术检测外周血中淋巴细胞所表达的RORγt mRNA的相对量,流式细胞仪检测外周血中Th17细胞的比例,ELISA法检测血浆中IL-17的水平。结果 RORγt mRNA在急性肺损伤组的表达水平(0.56±0.08)高于对照组(0.19±0.05),差异显著(P<0.01)。Th17细胞在急性肺损伤组的比例(32.60±5.51)%高于对照组(12.29±1.71)%,差异有统计学意义(P<0.01)。IL-17在急性肺损伤组的浓度(87.33±6.08)pg/ml明显高于对照组(52.71±3.19)pg/ml,P<0.01。结论 Th17细胞与急性肺损伤具有相关性,可能通过分泌IL-17来上调炎症反应,加重炎症反应的失控。 Objective To investigate the function of Th17 cell in ALI subjects.Methods Peripheral blood was abtained from 10 subjects with ALI and 10 control subjects.RORγt mRNA expression of lymphocytes was measured by RT-PCR,percentage of Th17 cell was detected by FCM,IL-17 in plasma was examined by ELISA.Results RORγt mRNA level of ALI group(0.56±0.08)was higher than that of control group(0.19±0.05)significantly(P0.01),proportion of Th17 cell of ALI group(32.60±5.51)% is significantly higher than that of control group(12.29±1.71)%,(P0.01),to compared with IL-17 level of control group(52.71±3.19)pg/ml,there was significant increase in ALI group(87.33±6.08)pg/ml,P0.01.Conclusion The results demonstrated that Th17 cell is associated with ALI,and probably promotes the disturbance of inflammatory reaction by secreting IL-17.
