摘要
目的 探讨 α-二氢麦角隐亭 DHEC对脊髓损伤后由血管痉挛造成 2次神经损伤的保护作用。 方法 对 72只 Wistar大鼠行阻断腹主动脉 2 5 m in后再灌流 ,制备成缺血性脊髓损伤模型。大鼠分成 :α-二氢麦角隐亭、海得静、尼莫地平和盐水对照组 ,分别于药物治疗的 3d、7d、14d处死动物 ,进行 CNTF抗体的 ABC免疫组织化学染色反应。 结果 脊髓横切片的 CNTF免疫反应结果经图像分析和 SAS软件处理后表明 ,在综合对比中 ,3种药物与对照组相比都具有显著性差异 (P=0 .0 0 0 1) ;α-二氢麦角隐亭组与海得静组和尼莫地平组的比较也都具有显著性差异 (P=0 .0 0 0 1) ;而海得静组与尼莫地平组间的比较的差异无显著性 (P=0 .2 6 31)。 结论 α-二氢麦角隐亭、海得静、尼莫地平都有增强脊髓损伤后脊髓运动神经元的 CNTF免疫反应性 ,而 α-二氢麦角隐亭的作用又强于海得静和尼莫地平。
Objective\ To observe the effects of dihydrergocryptine(DHEC) on the acute ischemic trauma of spinal cord,for the pharmacological and clinical studies showed that the dihydroergocryptine is effective against the manifestations of cerebral metabolic and circulatory insufficiency. Methods\ The animal model of spinal cord ischemic trauma was made by following 25min occlusion and re\|perfusion of peritoneal aorta above renal artery in 72 Wistar rats.Rats were divided into 4 drug groups:the dihydroergocryptine,hydergine,nimodipine and salt contral groups.After 3\,7\,14days of administration,the rats were killed and the CNTF antibody's immunohistochemistry was performed. Results\ It is shown that number and relative data of CNTF immuno\|like reactive cells of spinal cord in 3 drug administration group rats were statistical different to control group( P =0 0001);while the data of DHEC group was also statistically different to the groups of hydergine and nimodipine( P =0 0001);However,there was no statistical different between hydergine and nimodipine groups( P =0 2631). Conclusion\ Dihydroergocryptine,hydergine,nimodipine effect on the expression of CNTF after the ischemis trauma of spinal cord in the rats,and dihydroergocryptine was more effective than that of hydergine,nimodipine.
