摘要
目的研究哺乳期暴露双酚A(bisphenol A,BPA)对初断乳雄性仔鼠睾丸结构及雌激素受体α(ERα)表达的影响及可能机制。方法将18只健康清洁级妊娠ICR母鼠随机分为6组,分别为高(50 mg/kg)、中(5 mg/kg)、低(0.01 mg/kg)剂量BPA染毒组和阳性对照[0.1 mg/kg的雌激素(E2)]组、阴性对照(玉米油)组、空白对照(未经任何处理)组,每组3只母鼠。母鼠自分娩后第2天至第21天(断乳),灌胃染毒。观察第22天仔鼠睾丸形态学改变,并测定仔鼠睾丸中不同区域曲细精管直径;分别采用免疫组化法和Western blot法测定睾丸内ERα蛋白的表达水平。结果高、低剂量BPA染毒组及阳性对照组中睾丸部分节段横切面曲细精管多呈向心性区域性聚集;且高、低剂量BPA染毒组与阳性对照组聚集区域曲细精管直径大于阴性对照组(P<0.05)。与阴性对照组比较,各剂量BPA染毒组仔鼠睾丸内ERα蛋白的表达水平均降低(P<0.05,P<0.01),且ERα的表达水平随着BPA染毒剂量的升高而增加。结论哺乳期接触一定剂量BPA可不同程度地改变子代小鼠睾丸曲细精管的排列方式及成熟度,降低睾丸组织内ERα蛋白的表达水平。
Objective To study the effects of lactational bisphenol A (BPA) exposure on the structure of testis and the expression of estrogen receptor α(ERα) of weanling male mice. Methods A total of 18 pregnant ICR healthy mice were randomly divided into six groups, high,moderate and low dose BPA groups (exposure doses were 50,5 and 0.01 mg/kg body weight), estrogen (E2) positive control group (exposure dose was 0.1 mg/kg body weight),corn oil negative control group and blank group(without treatment). The treatment was conducted after delivery, by gavage to maternal mice during postnatal days (PND) 2-21. The morphology changes of testis and the diameter of rat testis seminiferous tubular in different regions were determined, and immunohistochemistry and western blot were used to determine the expression of ERα. Results The seminiferous tubule showed regional aggregation and seminiferous tubules lumen increased significantly (P〈0.05) in the outer area of testis transverse of the high-dose,low-dose BPA groups. The average diameter of seminiferous tubules in the regional aggregation arrangement was longer than that of all seminiferous tubules from some testis transverse in negative group (P〈 0.05). The expressions of ERα were decreased(P〈0.01 ,P〈0.05) in the testes of all BPA-treated groups,which were negatively correlated with BPA doses. Conclusion During lactational stage,exposure to a certain dose of BPA can influence the normal development of testicular cells, interfere with the arrangement and maturity of the seminiferous tubules and decrease the expression of ERα in the testicular tissue of male mice.
