摘要
目的:观察与探讨芪冬活血饮对急性肺损伤(ALI)大鼠肺表面活性蛋白A(SP-A)在肺与大肠等组织表达影响,并且对SP-A在肺与大肠表达的相关性进行分析,从而为中医"肺与大肠相表里"理论提供实验依据。方法:SD大鼠60只,随机分为6组,采用气道内滴入脂多糖(LPS)制备ALI模型,各组均分别应用相应药物进行干预。所有大鼠均在造模后24h处死采集标本,采用ELISA和免疫组化法检测各组织SP-A表达,并记录肺湿干重比及进行HE染色光镜下观察肺损伤程度。结果:造模后24h,肺、大肠、支气管肺泡灌洗液(BALF)及血清中SP-A表达均下降。芪冬活血饮可以显著促进肺组织及血清中SP-A含量(P<0.05),减轻肺水肿及肺组织病理学损害。芪冬活血饮同大黄合用,可以促进ALI大鼠肺和大肠组织SP-A表达,提高BALF及血清中SP-A含量,减轻肺水肿及肺组织病理学损害。通过相关性分析,发现肺组织SP-A表达同BALF、大肠及血清表达呈正相关,但与大肠相关最密切(r=0.615)。结论:芪冬活血饮可以促进肺组织、大肠、BALF及血清中SP-A表达,减肺水肿,改善肺组织病理学损害,并且同大黄具有协同作用;经相关性分析,肺组织SP-A表达与大肠相关最密切。
Objective: To investigate the effects of Qidong Huoxue Yin on expression of pulmonary surfactant protein A in lung and large intestine tissue in LPS induced acute lung injury rats. And to analyze the correlation of pulmonary surfactant protein A expression in lung and large intestine, in order to provide scientific basis for traditional Chinese medicine theory of 'the lung and large intestine have exterior-interior relationship'. Methods: Rat acute lung injury was established by intratracheally LPS instillation. 60 SD rats were randomly divided into six groups. Qidong Huoxue Yin and rhubarb alone or in combination was administrated intragastrically three times before and after LPS induction with dexamethasone and normal saline as positive and negative controls. Specimens were obtained 24h after LPS treatment. SP-A were detected by ELISA and Immunohistochemical stain. And lung wet-dry ratio and HE staining were used to examine lung edema and pathological destruction (changes). Results: SP-A in lung, large intestine, BALF and serum was significantly decreased 24h after LPS treatment. The SP-A reduction in lung and serum caused by LPS could be restored by administration of Qidong Huoxue Yin or rhubarb (P〈0.05), the increases of SP-A in BALF and large intestine were also observed, but the difference wasn't statistically significant. The pulmonary edema and histopathological damages were improved. Moreover, a synergetic effect was observed when Qidong Huoxue Yin and rhubarb were used together in increasing SP-A in lung, large intestine, serum and BALF, relieving pulmonary edema and histopathological damages. By correlative analysis, the expression of SP-A in lung was positively correlate with that in large intestine, serum and bronchoalveolar lavage fluid, but the correlation with large intestine is the most closely (correlation coefficient is r=0.615). Conclusion: Qidong Huoxue Yin could prevent SP-A decease in lung, large intestine, BALF and serum, reduce the pulmonary edema and histopathological damages induced by LPS in rat, and Qidong Huoxue Yin displayed a synergetic effect when used with rhubarb together. By correlative analysis, the correlation between the lung and large intestine is the most close.
