摘要
以芦丁为对照品、总黄酮提取得率为指标,采用正交试验法,考察提取温度、超声时间、乙醇浓度、料液比、提取次数等因素对金樱子中总黄酮提取得率的影响。结果显示,提取金樱子中总黄酮的最佳工艺条件为:乙醇浓度60%,料液比1∶20,提取温度70℃,超声时间60 min,提取3次;各因素对金樱子中总黄酮提取得率的影响顺序为乙醇浓度>料液比>提取温度>超声时间。在最佳工艺条件下,金樱子中总黄酮的提取得率为0.498%,约等于正交设计中的最高提取得率,表明通过正交试验研究达到了优化目的,优选的最佳提取工艺条件稳定可靠。
Taking Rutin as a reference substance, extraction ratio of flavonoids as a index, surveying how the extraction temperature, ultrasonic time, ethanol concentration, solid-liquld ratio, extract times and other factors affect the extraction ration of flavonoids by using the method of orthogonal test. The results showed that the best processing conditions for extracting ilavonoids in Rosa laevigata Mickx were: ethanol concentration 60%, solid-liquid ratio of 1:20, extracting temperature of 70~C, ultrasonic time of 60 min, extracting 3 times, the various factors which affect extracting ratio in a order of ethanol concentration 〉 solid-liquid ratio 〉 extracting temperature 〉 ultrasonic time. Under the best processing condition, it got a extraction ratio of 0.498% for flavonoids in R. laevigata Mickx, which was almost equal to the highest extraction ratio of orthogonal test. It indicates that using the method of orthogonal test reached the goal of optima, and the best extracting processing was reliable and stable.
