3个菊芋品种的ISSR引物筛选及分子鉴别 被引量:5

摘要 采用ISSR分子标记对3个菊芋品种进行了分析,从100条ISSR引物中筛选出16条多态性好、稳定性高的引物对3个菊芋品种的DNA序列进行了PCR扩增,共扩增出160个位点,多态性位点132个,比率为82.5%;建立了3个菊芋品种的分子识别卡,用2条引物的9个多态性位点即可将其分开;3个品种间遗传距离在0.44~0.86之间,平均为0.3,表明3个菊芋品种具有不同的亲缘关系;本研究为菊芋品种的鉴别及选育提供了理论依据。 Genetic differences and kindredship among 3 samples in Helianthus tuberosus L. were studied by adopting ISSR markers. 16 out of 100 ISSR primers were screened out and 160 ISSR markers were amplified from these clones with 132 polymorphie (percentage of polymor- phie bands was 82.5% ). The value of Neips genetic similarity (GS) indexes of 3 euhivam based on the ISSR data varied from 0.44 to 0. 86, with an average of 0.3, suggesting that the phylogenetie relationships of three Helianthus tuberosus L. were different. Molecular identifi- cation card was established to distinguish the three Helianthus tuberosus L. This study would provided information for cultlvar identification of Helianthus tuberosus L.
