摘要
目的:测定番荔枝子的醇提物与石油醚萃取物总内酯含量;观察番荔枝子醇提物及其石油醚萃取物的急性毒性,评价其安全性。方法:渗漉法制备番荔枝子醇提物,并从醇提物中萃取得到石油醚部位,对醇提物及石油醚萃取物同时进行急性毒性试验,比较LD50值。同时,测定醇提物及石油醚萃取物的番荔枝总内酯含量。结果:番荔枝子醇提物的LD50值为(1.04±0.18)g.kg-1[折合成生药量为(4.34±0.75)g.kg-1],石油醚萃取物LD50值为(2.53±0.39)g.kg-1[折合成生药量为(14.10±2.17)g.kg-1],而醇提物的番荔枝总内酯含量为12.00%,石油醚萃取物的番荔枝总内酯含量为9.84%,且石油醚中番荔枝总内酯量占整个醇提物中总内酯量的61.36%。结论:番荔枝子石油醚萃取物中含低极性番荔枝内酯较多,其毒性也降低。
Objective: The lactone content of ethanol extract and petrolum ether extract of A nnona squamosa semen were determined. To observe the toxicity of 95% ethanol extract and its petroleum ether portion from the seeds of A rmona squamosa L, and evaluate its safety. Methods: The LD50 of the 95% ethanol extract and its petroleum ether portion were determined and compared by taking acute toxicity test on mouse. Besides, the contents of total annonaceous acetogenins from each extract were determined by UV-VIS methods. Results: The LD50 was 1.04±10.18 g·kg^-1 (4.34±0.75g·kg^-1 as crude seeds) for the 95% ethanol extract, while 2.53±0.39 g·kg^-1 (14.10±12.17 g·kg^-1 as crude seeds) for the petroleum ether portion. The content of total annonaceous acetogenins was 12% in the 95% ethanol extract, while 9,84% in the petroleum ether portion. Besides, 61.36% of the total annonaceous acetogenins in the 95% ethanol extract was distributed in the petroleum ether portion. Conclusion: The toxicity of the petroleum ether portion is weaker, which contains more low polarity annonaceous acetogenins.
