柚皮苷调控心肌核因子NF-κB炎症信号通路对糖尿病心肌病大鼠防治作用 被引量:18

摘要 目的:探讨柚皮苷调控心肌核因子NF-κB炎症信号通路对糖尿病心肌病大鼠防治作用。方法:高糖高脂喂养和一次性腹腔注射低剂量STZ建立糖尿病心肌病大鼠模型,分为模型对照组和柚皮苷高、中、低剂量治疗组,另设正常对照组。免疫组织化学法测定心肌组织NF-κB蛋白的表达,放射免疫分析法分别检测外周血TNF-α、IL-6、IL-1β含量,透射电镜观察心肌组织超微结构。结果:与正常对照组比较,糖尿病心肌病大鼠心肌组织核因子NF-κB蛋白以及外周血TNF-α、IL-6、IL-1β表达明显增加(均P<0.05),糖脂代谢紊乱,心肌超微结构损伤明显,而在高、中剂量柚皮苷治疗组则明显受到不同程度改善,并呈剂量依赖性。结论:柚皮苷呈剂量依赖性对糖尿病心肌病具有明显的防治作用,其可能机制是有效地改善心肌糖脂代谢,下调了转录因子NF-κB的表达,继而抑制了NF-κB通路的激活。 Objective:To investigate preventive effect of naringin on cardiac NF-KB inflammatory signal pathway in diabetic cardiomyopathy rats. Methods:Diabetes cardiomyopathy rat model was established by High sugar and fat feed and one-time intraperito- neal injection of low dose STZ were divided into model control(MC) group and naringin in high, middele and lower dose treatment groups, the other set of normal control(NC) groupl The expression of NF-KB on myocardium was examined by immunohistochemistry and the levels of TNF-α, IL-6, IL-115 in serum were examined by radio immunoassay, myocardial uhrastructure was examined through e- lectron microscopy. Results:Compared with NC group, the expression of NF-KB on myocardium and the level of TNF-α, IL-6, IL-113 in serum were significantly increased in MC group ( P 〈 O. 05 ), and glucolipid metabolism disorders, myocardial uhrastructure injured were obvious significantly in MC group. The effect of the naringin in high, middele dose treatment group was better than that in the MC group, and with the dose dependent. Conclusion:Naringin can prevent and treat diabetic cardiomyopathy, improve myocardial glu- colipid metabolism, down-regulate of transcription factors NF-KB, and then to suppress the activation of cardiac NF-KB inflammatory signal pathway.
