摘要
目的 探讨柯萨奇B3病毒 (CoxsackievirusB3,CVB3)对膜离子通道及离子交换载体的影响 ,了解病毒感染导致细胞内钙超载的原因。方法 酶灌注法获得单个心肌细胞后用光聚焦显微镜和钙离子荧光探针 (Fluo 3/AM)检测CVB3感染对心肌细胞内游离钙离子浓度的影响及利用模片钳全细胞电流记录技术观察CVB3对L型钙通道电流、钠通道电流和钠钙交换电流的影响。结果 CVB3感染使细胞内的游离钙浓度从 5 1nmol/L± 44nmol/L增加到 339nmol/L± 2 0 8nmol/L ,使L型钙通道电流从 4.13pA/pF± 0 .75pA/pF增加到 4.9pA/pF± 1.4pA/pF ,使内向钠钙交交换电流从 - 0 .46pA/pF± 0 .2 1pA/pF降低到 - 0 .2 8pA/pF± 0 .16pA/pF ,并使钠钙交换电流的逆转电位从 - 2 9.4mV±7.0mV移向 - 41.2mV± 14mV。结论 CVB3增加L型钙通道电流 ,抑制内向钠钙交换电流是感染细胞内钙离子增多的一个方面。可能是病毒感染后细胞损伤和产生异常电活动的原因。
Objective To investigate the effect of coxsackievirus B3 (CVB3) on intracellular calcium, L type voltage dependent calcium channel(VDCC) and Na +/Ca 2+ exchange (NCE) in adult rat cardiomyocytes.Methods Rat hearts were perfused with collagenase to gain single cardiomyocyte .The intracellular free calcium concentration of the cardio myocytes were detected by laser cofocal microscopy with calcium indicator, Fluo 3 acetoxymehyl ester; and the currents of VDCC, Na + and NCE were recorded by patch clamp techniques.Results The free intracellular calcium concentration increased from (51±43) nmol/L in normal cardiomyocytes to (339±208) nmol/L in CVB3 infected cardiomyocytes. The VDCC current increased from (4.13±0.75) pA/pF to (4.9±1.4) pA/pF after CVB3 infection, and the inward NEC current decreased from (-0.46±0.21)pA/pF to (-0.28±0.16)pA/pF; besides, the reverse potential of NCE was shifted from (-29.4±7.0)mV to (-41±14)mV with CVB3 infection.Conclusion The increase of L type VDCC current and decrease of inward NCE current with shift of NCE reverse potential may partly attribute to intracellular free calcium accumulation; also, it may be the cause of cell damage and the occurrence of abnormal electric activities induced by CVB3 infection.
