高效液相色谱法同时测定银翘散中绿原酸、连翘苷、牛蒡苷、甘草苷、甘草酸的含量 被引量:20

摘要 目的建立同时测定银翘散中绿原酸、连翘苷、牛蒡苷、甘草苷、甘草酸含量的方法。方法采用高效液相色谱法,以绿原酸、连翘苷、牛蒡苷、甘草苷、甘草酸为对照,色谱柱为Kromasil C18柱(4.6 mm×250 mm,5μm),乙腈-0.4%磷酸溶液梯度洗脱,流速1.0 mL·min-1,检测波长230 nm,柱温为30℃。结果绿原酸、连翘苷、牛蒡苷、甘草苷、甘草酸分别在0.039 4~0.788、0.020 5~0.410、0.068~1.36、0.034 8~0.697和0.037 6~0.752μg内呈良好的线性关系,相关系数均大于0.999 6,平均回收率分别为101.3%(RSD=1.5%)、99.5%(RSD=1.2%)、98.7%(RSD=1.9%)、96.7%(RSD=2.3%)和98.2%(RSD=2.0%)。结论该法快速简便、准确可靠、重复性好,可用于银翘散中绿原酸、连翘苷、牛蒡苷、甘草苷、甘草酸含量测定,为银翘散及其类方制剂的质量评价提供研究基础。 OBJECTIVE To establish a method for the determination of chlorogenic acid, phillyrin, arctiin, liqniritin and glycyrrhizin in Yinqiao Powder. METHODS High performance liquid chromatography was performed on a Kromasil Clscolumn (4. 6 mm · 250 mm, 5 μm) at 30 ℃. The mobile phase was acetonitrile-0.4% phosphoric acid solution with gradient elution. The flow rate was 1.0 mL · min^-1 and the detection wavelength was set at 230 nm. RESULTS Chlorogenic acid, phillyrin, arctiin, liquiritin and glycyrrhizin had good liner relationship in the ranges of 0. 039 4 -0. 788, 0. 020 5 -0. 410, 0. 068 - 1.36, 0. 034 8 -0. 697 and 0. 037 6 -0. 752 μg, respectively. The correlation coefficients were all more than 0. 999 6. The average recoveries were 101.3% ( RSD = 1.5%), 99. 5% (RSD=l.2%), 98.7% (RSD=l.9%), 96. 7% (RSD=2.3%) and 98.2% (RSD=2.0%), respectively. CONCLUSION This method is convenient, rapid, accurate, reliable and has good reproducibility, which can be used for the determination of chlorogenic acid, phillyrin, arctiin, liquiritin and glycyrrhizin in Yinqiao Powder. This study provides scientific basis for the quality evaluation of Yinqiao Powder and preparations of the same kind.
