摘要
目的:探讨阿尔茨海默病模型大鼠海马CREB的变化以及电针对其的影响。方法:将40只SD大鼠按随机数字表分为4组,即正常组、假手术组、模型组、电针组。采用双侧海马一次性注射聚集态Aβ25~35制备AD大鼠模型,电针组选取双侧"肾俞"穴、双侧"内关"穴和"大椎"穴,接G6805-Ⅱ型电针治疗仪,选用2 Hz连续波,强度为1 mA,通电20 min,每天1次,每周6次,共治疗2周。治疗结束后,采用免疫组化法检测各组大鼠海马CREB表达的变化,并进一步分析电针对其的影响。结果:经图像分析,与假手术组比较,模型组大鼠海马CA1、CA3区CREB的表达明显降低,有显著性差异(P<0.01);电针组大鼠海马CA1区CREB的表达与模型组比较有显著提高(P<0.05),而CA3区CREB的表达虽有升高的趋势,但无统计学意义(P>0.05)。结论:电针可能通过增强海马CREB的表达,改善LTP,提高AD大鼠的学习记忆能力。
Objective:To explore the changes of cAMP - response element binding protein in the hippocampus of rats with Alzheimer's Disease and observe the effect of electro -acupuncture therapy on the hippocampus. Methods:Male SD rats in number 40 were randomly divided into 4 groups in average, the normal group, the sham group, the model group, and the EA group. By utili-zing condensed Aβ25 - 35 in one -time injection of bilateral hippoeampus to induce AD model rats, we selected bilat-erahn Shen- shu(BL23) ,bilat- cralm Neiguan(PC6) and Dazhui (GV14)acupoints in EA group. G6805 - Ⅱ type of electroaeupuneture treatment device was connected to needles, and then we selected the frequency of 2 Hz eontinus wave ( CW), the intensity of 1 mA. The therapy last 20 rains, qd,6 times a week,a total of 2 weeks treatment. The expression of CREB protein in hippocampus was assayed by immunohistochemistry. Results:Compared with the sham group,the expression of CREB in hippocampus CA1 ,CA3 zone of the model group was lower, and the difference was significant(P 〈 0. 01 ) ;compared with the model group, the expression of CREB in CA1 zone of the EA group was significantly inereased(P 〈 0. 05 ). The expression of CREB in CA3 zone of the EA group was increased,but there was no statistical significanee(P 〉0. 05). Conclusion:Its mechanism may be achieved through the following mean : increasing the expression of CREB in the hippocampus, enhancing LTP, in order to improve the learning - memo- ry capacity of rats with AD.
