摘要
利用Placket-Burman设计和响应面法对产3-羟基丁酮Bacillus subtilis SF4-3的发酵培养基进行优化,首先通过Placket-Burman设计筛选出影响较大的3个因素,分别为酵母膏、玉米浆和尿素,然后进行最陡爬坡实验逼近最佳响应面区域,最后根据Box-Benhnken的中心组合设计原理,进一步进行响应面分析实验,得到3因素的最佳浓度。在优化培养基下,3-羟基丁酮的产量达51.2g/L,比优化前提高了51%。
The aim of this work was to optimize the culture component for acetoin produced by Bacillus subtilis SF4-3 using Plackett-Burman (PB) design and response surface methodology. Firstly, Placket- Burman was applied to screen three main factors, including yeast extract, corn steep liquor and urea. The path of steepest ascent was used to approach the optimal region of the fermentation conditions subsequently. Then, the concentrations of the three factors were further optimized by Box-Behnken and response surface analysis, the optimized composition of medium was obtained. Under the optimized conditions, the concentration of acetoin could reach 51.2 g/L, which increased by 51% compared with the preliminary culture.
