摘要
目的:探讨葎草中总黄酮含量的测定方法。方法:以芦丁为对照品,采用超声提取法提取总黄酮,用亚硝酸钠-氯化铝-氢氧化钠显色法,最佳显色条件:亚硝酸钠质量浓度5%、氯化铝质量浓度10%、氢氧化钠质量浓度4%。通过紫外可见分光光度计测定律草中总黄酮的含量。结果:检测波长为510 nm,芦丁在浓度0.024 1~0.064 0 g·L-1范围内呈良好的线性关系,回归方程:y=0.081 7x-0.001 8(R2=0.999 5),1 h内基本稳定(RSD=0.364%),重复性较好(RSD=0.600%),加样回收率为99.46%(RSD=1.685%)。结论:紫外可见分光光度计测定葎草中总黄酮的含量,方法简便、快捷,结果准确、可靠。
Objective:To investigate the Humulus scandens content determination of total flavone. Methods:Using rutin as control sample, using ultrasonic extraction method to extract total flavonoids, with sodium nitrite-aluminum chloride-sodium hydroxide colorimetry, optimum chromogenic conditions:sodium nitrite concentration 5%, aluminum chloride concentration10%, sodium hydroxide concentration 4%. Through ultraviolet visible spectrophotometric determination of total flavone content. Results:The detection wavelength was 510 nm,rutin at a concentration of 0. 024 1 -0. 064 0 g · L^(-1) range is a good linear relationship, the regression equation:y = 0. 081 7x 0. 001 8(R2 = 0. 999 5 ), 1H ( RSD = 0. 364% ) basic stability, good reproducibility ( RSD = 0. 600% ), sample recovery rate was 99. 46% (RSD = 1. 685% ). Conclusion: UV visible spectrophotometric determination of total flavone content in Humulus scandens, method is simple, fast, accurate, and reliable.
