痛泻要方对内脏高敏性大鼠结肠MC活化与5-HT相关性影响的研究 被引量:12

摘要 目的:通过对内脏高敏性大鼠结肠MC密度、5-HT表达及二者相关性的影响,研讨痛泻要方对内脏高敏性大鼠结肠MC活化的干预作用。方法:采用乳鼠结肠刺激方法复制内脏高敏性大鼠IBS模型,以免疫组织化学方法和甲苯胺蓝改良法进行结肠双重复染法,观察痛泻要方对结肠组织MC形态学、密度的影响,及其与5-HT表达之间关系,并观察痛泻要方对其二者相关性的影响。结果:经乳鼠结肠刺激方法复制内脏高敏性模型后,结肠中MC颗粒增多,MC周边5-HT表达增强,二者有明显相关性,经痛泻要方干预后,结肠中MC形态改善、数量降低,5-HT表达降低,对二者相关性具有明显调节作用。结论:痛泻要方可改善内脏高敏性大鼠肠道MC活化程度,并抑制MC释放5-HT,从而降低5-HT表达。 Objective By studying on the effect on density of MC,the expression of 5-HT in intestinal and their relationship,to investigate the effect of Tongxie Yaofang on MC activation in visceral hypersensitivity rat colon.Methods:Establish IBS model of visceral hypersensitivity rats by milk mice colon irritation,to study the effect of Tongxie Yaofang on morphology and density of MC in colon,the expression of 5-HT,and the effect of Tongxie Yaofang on their correlation by double counterstaining method that immunohistochemical method and toluidine blue staining method.Results:After the model was established,particles in MC were increased,the expression of 5-HT around MC was enhanced,and there was an obvious correlation.After intervened by Tongxie Yaofang,the morphology of MC was improved,the quantity of MC was decreased and the expression of 5-HT was reduced.Its effect on their correlation had significantly changed.Conclusion: Tongxie Yaofang can improve degree of MC activation on visceral hypersensitivity rat in intestinal and inhibit MC release 5-HT to reduce the expression of 5-HT.
