人参皂苷Rh1诱导卵巢癌细胞凋亡及对piwi基因的表达影响 被引量:4

摘要 目的探索Rh1药物对SKOV3细胞生长的影响,评价Rh1药物对细胞piwi基因家族的表达调控进行研究。方法 MTT分析Rh1对细胞生长的作用,realtime PCR和western blotting检测Rh1对piwi基因的表达影响。结果 Rh1能够明显抑制SKOV3细胞的增殖,10-6,10-8,10-10和10-12M的Rh1处理组均能够明显抑制SKOV3细胞活力。piwil-1,piwil-2,piwil-3和piwil-4的mRNA和蛋白在Rh1处理后均有不同程度的表达下调。结论 Rh1可抑制人卵巢癌细胞SKOV3细胞增殖并使piwi基因家族表达降低。 [Objective] To detect Rhl effect on cell growth of SKOV3 and assess Rhl regulation of ex- pression of piwi gene family. [Methods] Rhl on cell growth was analyzed by MTT. The Rhl on expression of piwi gene were detected by RT-PCR and western blot. [Results] Rhl significantly increased SKOV3 via- bility. The mRNA and protein expression of piwil-1, piwil-2, piwil-3 and piwil-4 were decreased after Rhl treatment. Rhl could inhibit human ovarian cancer cells proliferation. 10^-6, 10^-8, 10^-10 and 10^-12 M Rhl treat- ment groups could depress expression of piwil-1, piwil-2, piwil-3 and piwil-4. [Conclusion] Rhl can inhibit human ovarian cancer cells proliferation and decrease the expression of piwi gene.
