摘要
目的探讨一种效果较好的水中病毒富集方法,为水中病毒的检测提供依据。方法向高压灭菌处理的水样中接种已知滴度的脊髓灰质炎Ⅰ型病毒,采用载阳电荷滤料法、膜吸附-洗脱法Ⅰ、膜吸附-洗脱法Ⅱ对水样分别进行病毒富集后,利用荧光定量RT-PCR技术检测并计算回收率。结果当水中病毒载量为7×102~7×105pfu/L时,载阳电荷滤料法的病毒回收率为92%~94%;2种膜吸附-洗脱法的回收率为72%~77%。结论载阳电荷滤料法富集水中病毒的效果好于2种膜吸附-洗脱法。
Objective To study an effective method for concentration of viruses from water,so as to provide a basis for detection of virus in the water.Methods To the autoclave treated water samples inoculate with known titer of poliovirus type Ⅰ,then the viruses in the water samples were concentrated by using electropositive material method,membrane adsorption-elution method Ⅰ,and membrane adsorption-elution method Ⅱ respectively.And the viruses were detected by using fluorescent quantitative RT-PCR.Results The virus recovery rate was 92%~94% for the electropositive material method,72%~77% for the two membrane adsorption-elution methods when the titer of virus was(7×102~7×105) pfu/L.Conclusion The effect of virus concentration is better in the electropositive material method than in the other two membrane adsorption-elution methods.
