摘要
目的:研究靶向NBS1的RNA干扰对鼻咽癌CNE-2细胞的放射增敏作用。方法:靶向NBS1的小干扰RNA转染鼻咽癌CNE-2细胞48小时后,4Gy剂量X射线照射。照射后24小时,流式细胞仪分析鼻咽癌细胞凋亡率。结果:X线照射+RNAi-NBS1组的细胞凋亡率显著高于单独X线照射组和X线照射+RNAi-阴性对照组(P<0.01)。结论:靶向NBS1的RNA干扰在鼻咽癌CNE-2细胞中有显著的放射增敏作用,诱导细胞凋亡是其放射增敏的重要机制之一。
Objective:To investigate the radiosensitizing effect of RNAi-NBS1 on the nasopharyngeal carcinoma CNE-2 cells.Methods:The nasopharyngeal carcinoma CNE-2 cells had been transfected with siRNA targeting NBS1 for 48h before X-ray irradiation at 4 Gy.Twenty four hours after irradiation,flow cytometry was used to analyze the apoptosis of CNE-2 cells.Results: The percentage of apoptotic cells was significantly increased in irradiation plus RNAi-NBS1 group when compared with the irradiation alone group and irradiation plus RNAi-control group(P0.01).Conclusion: The RNAi targeting NBS1 has a radiosensitizing effect on nasopharyngeal carcinoma CNE-2 cells,and this mechanism may be closely associated with the increase of the sensitivity of CNE-2 cells to X-ray irradiation-induced apoptosis.
