麻杏石甘汤对流感病毒感染小鼠肺部炎症与神经氨酸酶活性影响的研究 被引量:38

Lung inflammation and neuraminidase activity of Maxing Shigan Decoction on mice which were infected by influenza virus

摘要 目的:进一步探讨麻杏石甘汤抗流感病毒的作用及机制。方法:①经鼻腔接种建立小鼠流感病毒肺炎模型。实验设空白对照组、模型对照组、奥司他韦组和麻杏石甘汤组,灌胃给药7d后,观检肺指数和肺组织病理变化。②以2-(4-methylumbellifery1)-α-D-N-acetylneuraminic acid为底物,检测麻杏石甘汤全方与拆方配伍组合对流感病毒神经氨酸酶活性的影响。结果:与空白对照组比较,模型对照组肺组织大量淋巴细胞浸润,肺指数明显增加(P<0.01)。与模型对照组比较,全方组肺组织病理变化得到改善,肺指数明显降低(P<0.01)。与模型对照组比较,全方组、麻黄组神经氨酸酶活性依次下降53.4%、75.4%;麻黄+杏仁、麻黄+石膏、麻黄+甘草、麻黄+杏仁+石膏、麻黄+石膏+甘草、麻黄+杏仁+甘草、麻黄+杏仁+石膏+甘草等7个含麻黄的配伍组合神经氨酸酶活性依次下降69.9%、75.5%、49.4%、67.1%、43.1%、46.7%、53.7%。杏仁+石膏,杏仁+甘草,石膏+甘草,杏仁+石膏+甘草等4个不含麻黄的配伍组合神经氨酸酶活性依次下降13.5%、2.6%、3.3%、-1.2%。结论:麻杏石甘汤能明显减轻流感病毒感染小鼠肺部炎症反应,降低肺指数,其机制可能与其抑制神经氨酸酶活性,阻止病毒增殖有关。麻黄在该方抗流感病毒作用中有重要地位。 Objective: To further study the effect mechanism of Maxing Shigan Decoction in against influenza virus.Methods: ①Use nasal inoculation to establish mouse pulmonary infection model animals by influenza virus.Set up normal control group,model control group,Oseltamivir group and Maxing Shigan Decoction(MXSGD) group.After 7 days of drug or normal saline gavages,observing the lung index and pathological changes of lung tissue.②Using 2-(4-methylumbellifery1)-α-D-Nacetylneuraminic acid as a substrate,detecting the influenza virus neuraminidase activity of Maxing Shigan whole decoction and Maxing Shigan decoction with different concerted application.Results: ①Compared with the normal control group,lungs of model group have massive lymphocytic infiltration,the lung index was significantly increased(P0.01).Compared with the model group,pulmonary pathologic of lungs changes were significant improving in Maxing Shigan whole Decoction group,the lung index was significantly increased(P0.01).②Compared with the model group,the neuraminidase activity of Maxing Shigan Decoction group and herba ephedrae group was decreased by 53.4% and 75.4% respectively;The neuraminidase activity of herba ephedrae with almond,herba ephedrae with gypsum,herba ephedrae with glycyrrhizae,herba ephedrae with almond and gypsum,herba ephedrae with gypsum and glycyrrhizae,herba ephedrae with almond and glycyrrhizae,herba ephedrae and almond with gypsum and glycyrrhizae was decreased by 69.9%,75.5%,49.4%,67.1%,43.1%,46.7%,53.7%,respectively.While the neuraminidase activity of 4 concerted application groups with no herba ephedrae-almond and gypsum,almond and glycyrrhizae,gypsum and glycyrrhizae,almonds and gypsum and glycyrrhizae was decreased by 13.5%,2.6%,3.3%,-1.2%,respectively.Conclusion: Maxing Shigan Decoction could obviously alleviate lung inflammatory in influenza virus infected mice and reduce lung weight index,its mechanism may relate to inhibit the activity of influenza virus neuraminidase and prevent the proliferation of influenza virus.Herba ephedrae is playing a significant role in against the influenza virus.
