摘要
为了研究增强型绿色荧光蛋白(enhanced green fluorescent protein,EGFP)在上皮细胞中的表达情况,试验采用脂质体法介导真核表达质粒EGFP-C1转染PK15细胞和JEG-3细胞,分别在转染的24小时和48小时观察荧光的表达情况。结果表明:在荧光显微镜下,EGFP蛋白在PK15细胞和JEG-3细胞中均得到表达,相同条件下PK15细胞的转染效率高于JEG-3细胞,且在JEG-3细胞中EGFP均一表达,而在部分PK15细胞中EGFP表达主要聚集于细胞核区域,细胞状态对亚细胞定位存在重要的影响。
To study the expression of enhanced green fluorescent protein (EGFP)in epithelial cells, the eukaryotic expression plasmid EGFP - C1 was transfected into PK15 cells and JEG - 3 cells by liposome - mediated induction, and the expressions of fluorescence were observed when transfeeted for 24 hours and 48 hours, respectively. The results showed that EGFP protein was expressed in both PK15 cells and JEG -3 cells under the fluorescence microscope. Under the same condition, the transfeetion efficiency of PK15 cells was higher than that of JEG - 3 cells, and the fluorescent protein of EGFP was homogeneously expressed in the JEG - 3 cells, hut the expression of EGFP mainly accumulated in the region of nucleus of some PK15 cells, and the cell state had an important impact on the suhcellular localization.
