摘要
以4个杏品种为试材进行离体培养,在含各品种萌发最适蔗糖、硼酸浓度的培养基上观察比较不同品种24.0h内花粉萌发及花粉管生长动态。结果表明,花粉萌发率随蔗糖浓度的增加呈先增后降的变化趋势,乔尔胖花粉萌发最适蔗糖浓度为15%,大尤佳、洛浦洪待克与特尔湾玉吕克则均为20%。乔尔胖、大尤佳、特尔湾玉吕克花粉萌发最适硼酸浓度为0.01%,高于此浓度萌发受抑制,洛浦洪待克花粉萌发率则随硼酸浓度的增加呈先升后降变化趋势,硼酸浓度为0.05%时萌发率最高。4个品种中,乔尔胖花粉在培养6.0h内大量萌发,花粉管快速生长时间延至8.0h,大尤佳花粉大量萌发时间为1.0~10.0h,而花粉管快速生长时间为1.0~6.0h,洛浦洪待克花粉在8.0h内大量萌发,花粉管则仅在4.0h内快速生长,特尔湾玉吕克花粉萌发与花粉管生长高峰均在离体培养后6.0h内。离体培养各时段各品种花粉萌发率与自身花粉管长度间存在极显著正相关关系。除特尔湾玉吕克外,大尤佳、乔尔胖、洛浦洪待克花粉萌发率依次降低,但花粉管长度排序则相反。不同杏品种花粉离体萌发需要适宜的蔗糖、硼酸浓度,离体培养过程花粉在前期大量萌发,快速生长,后期萌发与花粉管生长减缓甚至停滞,品种间花粉萌发生长速率存在差异。
4 apricot varieties were cultured in vitro,on the medium the suitable concentrations of sucrose, boric acid for different varieties pollen germination, the dynamic of pollen germination and pollen tube growth of different varieties were observed within 24.0 h after cultured in vitro. The results showed that pollen germination rate increased first and decreased afterwards with the sucrose concentration increasing, the optimum sucrose concentration for Qiaoerpang pollen germination was 15 ~, those of Dayoujia, Luopu- hongdaike and Teerwanyulvke all were 20~. The optimum concentration of boric acid for Qiaoerpang, Dayouiia and Teerwanyulvke germination were 0.01%, and then the higher concentration would inhibit the pollen germination, while pollen germination rate of Luopuhongdaike increased first and then decreased with the rise of boric acid concentration. Which boric acid concentration was 0.05 % it had the highest ger- mination rate. Among 4 varieties,pollen of Qiaoerpang germinated was in large quantities within 6.0 h af- ter cultured,and rapid growth of pollen tubes lasted to 8.0 h, a large number of pollens germination of Dayoujia during 1.0 -- 1 0.0 h, whilefastgrowingphaseofitspollentubeswerein 1.0 -- 6.0 h, a large numberof Luopuhongdaikepollens germinated within 8.0 h,but its pollen tubes grew rapidly only in 4.0 h,the peak of pollen germination and pollen tube growth of Teerwanyulvke both were within 6.0 h after cultured in vitro. In different periods during cultured in vitro there were significant positive correlations between pollen germination rate and pollen tube length of differend varieties. Except Teerwanyulvke, germination rate of pollen of Dayoujia, Qiaoerpang, and Luopuhongdaike were decreased successively, but the sequencing of pollen tube length was exactly the opposite. Pollen germination of different apricot varieties in vitro re- quired suitable concentration of sucrose, boric acid, a large number of pollens germinated, and pollen tube rapidly grew in the earlier stage of culture process, and in the late stage of culture process, the pollen ger- minated and pollen tubes grew both slowly even growth rate of the different varieties. stagnated, there were differences in pollen germination and
