摘要 目的探讨磁性纳米顺铂微粒热化疗抑制卵巢癌skov-3转移相关基因CD44v6、基质金属蛋白酶2(MMP-2)表达的作用。方法取对数生长期的人卵巢癌skov-3细胞株分为阴性对照组、裸药组、纳米药物组、磁热组和磁热化组。处理并体外培养后采用RT-PCR检测各组MMP-2和CD44v6的表达。结果磁热化组CD44v6、MMP-2 mRNA的表达较其余4组有明显降低(P<0.05)。结论在电磁场介导下的磁性纳米顺铂微粒可以显著抑制卵巢癌侵袭转移相关基因CD44v6、MMP-2的表达,可能有抑制卵巢癌的作用。 Objective To study the magnetic nano-microspheres heat chemotherapy with cisplatin inhibits metastasis-related gene expression of ovarian cancer. Methods The human ovarian cancer skov-3 cells in logarithmic growth phase were divided into five groups of control group, drug-naked group, llano drug group, magnetic heat group, and heating magnetic group. After cultured in vitro, the expressions of matrix metalloproteinase-2 (MMP-2) and CD44v6 were detected with RT-PCtL Results The expressions of MMP-2 and CD44v6 were significantly lower in magnetic heating group than those in the other 4 groups (P〈0.05). Conclusion Mediated in the electromagnetic field,the magnetic nano-particles of cisplatin can significantly inhibit the metastasis-related gene CD44v6 and MMP-2 expressions in ovarian cancer, which may have inhibitory effect on ovarian cancer.
