豚鼠颈内静脉置管模型的构建

摘要 目的构建豚鼠颈内静脉置管模型,解决豚鼠静脉多次、大剂量给药问题。方法 10只豚鼠麻醉后均在显微镜下切开颈内静脉,置入医用PE导管,术后7d每日换药并向导管内注射1mL肝素化的生理盐水(50U/mL),观察有无堵管、脱落现象以及伤口有无红肿、渗液,术后第7d抽取静脉血行血常规及"C"反应蛋白检查。结果①术后观察7d,10只豚鼠存活8只,2只豚鼠于第4d死亡,每日观察无脱管及堵管现象。②术后第1d2只豚鼠伤口有明显渗出,略肿胀,第2d渗出减轻,无肿胀,此后至第7d伤口无渗出及肿胀出现;③术前与术后7d血常规主要指标与"C"反应蛋白均无明显差异。结论成功构建豚鼠颈内静脉置管模型,可满足静脉多次、大剂量给药的需要。 Objective To develop a new method to cannulate internal jugular vein in guinea pig for continuous heavy dose medication.Methods 10 albino guinea pigs were cannulated via internal jugular vein under microscope with medical PE cannula after anesthesia.In the first week after the operation,the dressing of wound was changed,and 1 mL normal saline mixed heparin(50 U/mL) was injected into the cannula every day.Meantime,whether each cannula was blocked,fell off or not,and whether there was swelling and exudation in the wound or not were recorded.In the 7th day post-operation,Routine Blood Test and C-reactive protein were examined.Results During the operation experiment 10 guinea pigs were successfully cannulated.In the first week after the operation no cannula was blocked or fell off except that 2 guinea pigs died in the 4th day.In the first day 2 guinea pig had their wound exuded and slightly swelled and in the 2nd day the exudation was alleviated and swelling disappeared.From the 3rd day to 7th day no exudation and swelling occurred.There were no differences before and after 7th days the operation in complete blood count and C-reactive protein.Conclusion A new method to cannulate internal jugular vein in guinea pig is established and can be applied to continuous heavy dose medication experiment.
