摘要
将人参茎叶皂甙(GSLS)和油佐剂混合,然后再与口蹄疫病毒(FMDV)抗原乳化制成口蹄疫疫苗。将FMD疫苗皮下二次免疫小鼠后,检测血清特异性IgG水平、脾脏淋巴细胞增殖指数(SI),脾淋巴细胞IFN-γ和IL-4mRNA表达水平,并测定GSLS对疫苗黏滞性的影响。结果表明,用200μL的FMD疫苗免疫小鼠,显著促进小鼠产生以4μg组产生最高水平的IgG(P<0.05);与对照组比较,添加GSLS后脾淋巴细胞对ConA和LPS增殖指数,IFN-γ和IL-4mRNA表达也显著增高(P<0.05);吐温浓度为1.0%和1.5%时,添加GSLS能显著降低疫苗的黏滞性(P<0.05)。因此,在FMD疫苗中添加GSLS能提高机体的体液免疫和细胞免疫应答,降低疫苗黏滞性。
Mice were immunized with FMDV antigen in oil emulsion with GSLS. Blood samples were collected for the measurement of IgG level. Splenocytes were prepared for the detection of lymphocyte proliferation and mRNA expression of IFN-γ and IL-4 in vitro. The viscosity of oil emulsion vaccines were measured with a viseometer. Results showed that the highest IgG levels was observed in mice immunized with FMD emulsion vaccine containing 4 /2g of GSLS (P〈0. 05). GSLS significantly promoted the lymphocyte proliferation in response to ConA and LPS and mRNA expression of IFN-T and IL-4 (P〈0.05). Compared with control group,co-administration of GSLS (4 zg) significantly reduced the viscosity in vaccines containing polysorbate-80 of 1.0% and 1.5% (P〈0.05) ,respectively. Results suggest that GSLS could improve both humoral and cellular immune responses to FMDV in mice and reduce the viscosity of emulsion vaccine.
