摘要
在生理酸度(pH=7.4)条件下,采用紫外-可见光谱法、荧光光谱法及熔点实验,研究了芍药苷与小牛胸腺DNA的相互作用以及Zn2+、Ca2+和Mg2+的影响。结果表明:芍药苷主要以沟槽模式与DNA结合。Zn2+、Ca2+和Mg2+分别能不同程度影响芍药苷的吸收光谱和DNA-双苯并咪唑(Hoechst 33258)复合物的荧光强度。随着体系中Mg2+浓度的增加,DNA-芍药苷复合物的结合常数呈现出先增大后减小的趋势,增加Zn2+的量则使复合物的结合常数逐渐增大,Ca2+浓度的增大对复合物结合常数的影响类似且弱于Zn2+。3种金属离子对DNA-芍药苷结合的影响取决于金属离子与DNA碱基以及磷酸基团间的结合程度。
The interaction between paeoniflorin (PF) and calf thymus DNA and the effect of Zn2+,Ca2+ or Mg2+ on this interaction in physiological buffer(pH=7.4) were investigated by UV-Vis absorption, fluorescence spectroscopy and melting experiments. The results revealed that paeoniflorin bound to DNA mainly through groove binding. All the three metal ions were found to be able to form complex with paeoniflorin making a difference to the absorption peak intensity of the reaction systems and quenching the fluorescence intensity of the DNA-Hoechst 339,58 complex to different extents.
