镇肝熄风汤预处理对大脑中动脉梗死大鼠脑组织ET-1表达、TNF-α含量及髓过氧化酶活性的影响 被引量:8

摘要 目的:研究镇肝熄风汤预处理对大脑中动脉梗死(MCAO)大鼠脑组织的内皮素-1(ET-1)、肿瘤坏死因子-α(TNF-α)的表达水平以及髓过氧化物酶(myeloperoxidase,MPO)活性的影响。方法:采用线栓法制备大脑中动脉梗死大鼠模型。于MCAO术前14天给予不同剂量镇肝熄风汤灌胃。实时定量PCR检测干预后脑组织的ET-1的mRNA,免疫组化法检测ET-1蛋白的表达,ELISA法检测的TNF-α含量,分光光度法检测MPO活性。结果:与假手术组相比较,模型组大鼠脑组织ET-1、TNF-α含量显著性增加(P<0.01),MPO的活性明显增加(P<0.01)。镇肝熄风汤干预14d后,MACO大鼠脑组织ET-1的表达明显减少(P<0.01),TNF-α含量明显减少(P<0.01),MPO的活性明显减弱(P<0.01)。结论:镇肝熄风汤通过抑制ET-1、TNF-α表达及局部炎症反应可能是其防治缺血性脑卒中的主要生物学基础。 OKJECTIVE To study the effect of Zhengan Xifeng decoction (ZGXFD) pretreatment on ET-1 expression, TNF-α content and myeloperoxidase (MPO) activity in rat cerebral tissue of middle cerebral artery occlusion(MCAO). METHODS The model of focal cerebral ischemia in rats was established with the suture-occuluded method and and was used to intervene with ZGXFD before MCAO. The levels of ET-1 in cerebral tissue after ZGXFD intervention were detected by fluorescence quantitative RT-PCR and immunohistochemistry method. The content of TNF-α and the activity of MPO in cerebral tissue were de- tected by ELISA and spectrophotometry. RESULTS After intervention by ZGXFD, the expression of ET-1, the content of TNF-α and MPO activity were significantly reduced in cerebral tissue of MCAO(P〈0. 01). CONCLUSION ZGXFD could directly modulate the ET-1 and TNF -α expression, improve local inflammatory response.
