摘要
药物分子的立体化学决定了其生物活性,手性已成为药物研究的一个关键因素,生物技术在手性药物合成中具有重要意义,利用酶催化的相关性质,通过酶拆分外消旋体、酶法不对称合成等方法合成手性药物,采用定向进化技术、酶分子修饰、辅酶再生等方法对手性药物合成方法进行改进,该文对近些年来生物技术在手性药物合成中的应用情况进行了综述。
The molecular stereoehemistry of drugs determines its biological activity. Chiral drug research has become the key factor of drug research, and biological technology is of great significance in the synthesis of chiral drugs. By utilizing the related properties of enzyme catalysis, chiral drugs are synthesized through the enzymatic resolution of racemate and enzymic method of asymmetric synthesis. The synthetic method of chiral drugs is improved and modified by utilizing directed evolution technology, enzyme molecule modification and coenzyme regeneration method. This paper gives an overview of the application of biological technology in the synthesis of chiral drugs in recent years. Key words: chiral drugs ; biological technology ; synthesis ; overview
