摘要
钠/氢交换调节因子2(Na^+/H^+exchangerregulatoryfactor2,NHERF2)是由337个氨基酸组成的多功能连接蛋白。NHERF2通过其PDZ—I、PDZ—II和ERM结合结构域与多种蛋白质结合,对Cl^-、Na^+/Pi等离子转运进行调节,影响PI,K/Akt、Erk等生长信号途径及对细胞迁移进行调控。参与调节与电解质及液体运输失调相关的疾病、氯离子通道功能障碍相关疾病以及肿瘤等疾病的形成过程。文章通过对其分子结构、调控的细胞功能及其与疾病发生、发展的关系进行综述,从而为研究相关疾病的防治提供新线索。
NHERF2 ( Na +/H ^+ exchanger regulatory factor 2) is a multifunctional adapter pro- rein consisting of 337 amino acids. NHERF2 interacts with a variety of proteins through the PDZ-I, PDZ-II and ERM-binding domain. It can regulate CI-, Na+/ Pi ion transport, PI3K/Akt, Erk growth signaling pathways and cell migration. It is involved in development of electrolyte transport disorders, chloride channel dysfunction, cancers and other diseases. In this study, the molecular structure of NHERF2, its ability to cell function regulation and the relationship between disease de- velopment and NHERF2 expression are discussed so as to provide new clues to prevent and treat some diseases.
