The Functions of ISL1 in Tumorigenesis

摘要 胰岛因子1(Islet1,ISL1),又称胰岛素增强子结合蛋白1,以多效转录因子的身份在多种组织器官的发育及成熟中发挥作用.ISL1在心、胰腺、神经系统等组织器官的胚胎发育过程中发挥重要作用.Isl1基因敲除可导致小鼠心发育不全,4种胰岛内分泌细胞缺失,以及运动神经元分化、迁移障碍等.ISL1具有促进增殖、抑制凋亡的重要功能.近年的研究表明,ISL1在多种肿瘤发生中存在异常高表达.然而,其在肿瘤发生中的具体作用与机制尚未被阐明,有待进一步探索. ISL1 (insulin gene enhancer binding protein 1) is a LIM-homeodomain transcription factor. It is known to play important roles in the development and maturation of multiple organs, including heart, pancreas and nerve system. The Isll knockout mice have shown multi-defects of cardiac development, absence of 4 types of pancreatic endocrine cells, and abnormal differentiation and migration of motor neurons. ISL1 is able to promote cell proliferation and inhibit cell apoptosis in the heart, nerves and pancreas islets. Recently, the abnormal overexpression of ISL1 was found in various tumors, indicating that the pathologic function of ISL1 and the underlining mechanism in tumorigenesis is worthwhile to be investigated.
