摘要
研究了3种抗氧化剂(抗坏血酸、亚硫酸钠、柠檬酸)对3个大花蕙兰品种组织培养的影响。结果表明:在一定浓度下三者配合使用能明显抑制褐变的产生,在含有抗坏血酸0.125 g/L、亚硫酸钠0.125g/L、柠檬酸2.5g/L及BA 4 mg/L+NAA 0.5 mg/L+KT 0.5 mg/L的MS培养基上,3个品种的芽增殖系数分别为8.3、7.1和4.0,与对照相比差异达极显著水平;但高浓度抗氧化剂会对大花蕙兰产生伤害。
The tissue culture of 3 C. hybridium varieties was carried out at different concentrations of ascorbic acid,sodium sulfite and citric acid to study their inhibiting effects on the explant contami- nation and browning. The results showed that combined application of the 3 antioxidants at some con- centration could significantly inhibit the contamination and browning. When the explants were cul- tured on MS medium containing 0. 125 g/L ascorbic acid,0. 125 g/L sodium sulfite and 2.50 g/L citric acid as well as 4 mg/L BA,0.5 mg/L NAA and 0.5 mg/L KT,the bud multiplication coefficients of the 3 varieties could reach 8.3,7.1 and 4.0 respectively and significantly higher than those on the medium without the antioxidants,but the high-concentration antioxidants could injure C. hybridiurn.
