摘要
目的:通过观察泽泻提取物对自发性糖尿病(GK)大鼠Bmal1(Arntl)、Acsl5、Gpx1基因表达的影响,以期揭示泽泻降低血糖、血脂可能的基因基础从而为临床应用提供理论依据。方法:GK大鼠随机分为泽泻组(n=9)和空白组(n=9),泽泻组给予提取物0.1 mL(30 mg)每100 g大鼠体重灌胃,1 d2次(约为人用量30 g.d-1),空白组给予等体积生理盐水。给药30 d后,麻醉取肝脏,抽提总RNA,反转录后,定量RT-PCR分析各基因mRNA表达量。结果:泽泻提取物极显著抑制了Bmal1基因的高表达(P<0.001),而对Acsl5、Gpx1的表达无显著影响(P>0.05)。结论:泽泻降低血糖、血脂并用于糖尿病治疗的机制可能与其抑制Bmal1的高表达有关。泽泻对GK大鼠Bmal1、Acsl5、Gpx1基因的调控方式与中医"治未病"以及"治本"的理念相一致。
Objective: To observe the effect of rhizoma alismatis extract(RAE) on Bmall (Arntl), AcslS, Gpxl gene expressions in spontaneous diabetic Goto - Kakizaki rats (GK) so as to demonstrate the mechanism of RAE induced glucose&lipid -lowering effects which could provide new evidence for clinical application. Methods: GK rats were ran- domized into RAE treatment group( n = 9) compared with blank control (n = 9 ). RAE was administrated intragastricly (i. g. ) with an dosage of 0.1 mL(30 mg) per 100 g weight, twice a day, while equal amount of physiological saline was given in control group. After the treatment of 30 days, animals were anesthetized and livers were detached. Total RNAs were isolated from liver tissues and quantitative real - time PCR was performed after reverse transcription. Results : The mRNA expression of Bmall was significantly suppressed in RAE treated group (P 〈 0. 001 vs control) while there was no significant change in Acsl5 and Gpxl mRNA expression levels( P 〉 0.05 vs control). Conclusions: Our findings indicate that the glucose&lipid - lowering effects of RAE as well as its therapeutic benefits against diabetes might be related with its significant suppression to Bmall mRNA expression. The regulation of Bmall, Acsl5 and Gpxl induced by RAE were consistent with the theory of preventive treatment and attacking the root causes in traditional Chinese medicine.
