摘要
建立了高效液相色谱同时测定复方脑得生有效部位中葛根素、3'-甲氧基葛根素、芹糖基葛根素和大豆苷含量的分析方法。采用Hyspersil ODS-2色谱柱,以甲醇-水(25∶75)为流动相等度洗脱,流速为0.9mL/min,紫外检测波长为250 nm,柱温为25℃。4种化合物在20 min内均得到良好分离,在考察的浓度范围内线性良好(r≥0.999 7),精密度、稳定性和重复性实验的相对标准偏差(RSD)均小于2.2%,平均加标回收率为96%~102%。运用该方法对复方脑得生有效部位中的4种异黄酮进行同时测定,总含量在37.3%~37.8%之间。
A reversed-phase high performance liquid chromatography coupled with UV detection (RP- HPLC/UV) was developed and validated for the simultaneous determination of four isofla- vonoids, including puerarin( PU), 3-methoxy puerarin( MP), mirificin(MI) and daidzin( DA), in the bioactive fraction of Naodesheng prescription, i.e. a widely used herbal medicine (HM) in Chi- na mainly for treatment of cerebral arteriosclerosis, isehemic stroke, sequelae of cerebral hemorrhage and etc. The separation of four compounds was accomplished on a Hyspersil ODS - 2 column ( 250 mm×4.6 mm, 5 μm i. d. ) by using methanol -water(25 : 75 ) as mobile phase with isocratic elu- tion at a flow rate of 0.9 mL/min. The UV detection wavelength was set at 250 nm, and the column temperature was 25 ℃. The high resolution and reproducible peaks were achieved for four compounds in 20 min. All the calibration curves showed good linearities( r≥0. 999 7 ) over the test concentration range. The relative standard deviations (RSDs) of the method were less than 2. 2% for precision, stability and reproducibility assays, and the spiked recoveries were in the range of 96% - 102%. The total contents of the four isoflavonoids were determined to be in the range of 37.3% - 37.8% in the bioactive fraction of Naodesheng prescription.
