IGF-Ⅰ和IGF-ⅠR在结直肠癌组织中的表达及临床意义 被引量:2

EXPRESSION AND CLINICAL SIGNIFICANCE OF IGF-Ⅰ AND IGF-Ⅰ R IN COLORECTAL CANCER

摘要 目的:检测IGF-Ⅰ、IGF-ⅠR在结直肠癌及癌旁正常组织中蛋白水平的表达,分析其与临床病理学因素的关系,探讨其与结直肠癌的发生、浸润、分化、转移及预后的相关性,以便用于结直肠癌的早期诊断,预后判断及靶向治疗。方法:应用组织芯片及免疫组化技术,检测IGF-Ⅰ、IGF-ⅠR在结直肠癌组织、癌旁正常组织中蛋白水平的表达。结果:在结直肠癌组织中,IGF-I、IGF-ⅠR的蛋白水平阳性表达率分别为64.58%、58.33%,均显著高于癌旁正常组织中的16.67%、22.9%(P均为<0.05)。IGF-ⅠR表达水平与肿瘤的浸润程度、淋巴结转移、Dukes分期有关(P均为<0.05),而与其他病理因素无关(P>0.05),差异有统计学意义。结论:IGF-Ⅰ、IGF-ⅠR与结直肠癌的发生、发展中有关。IGF-ⅠR蛋白水平过表达与结直肠癌的浸润深度、Dukes分期有关(P<0.05),IGF-ⅠR对结直肠癌的早期诊断、预后判断及靶向治疗有重要意义。 Objective :To detect the different expression of Insulin-like growth factor-I and Insulin -like growth factor-I receptor in colorectal carcinoma and adjacent precancerous tissues, and analyze the relations among IGFs, genesis, infiltration, differentiation, metastasis and prognosis of colorectal cancer, and to clarify the clinical significance of IGFs. More information from our in vivo study will give benefits for early diagnosis and target therapy of colorectal cancer. Methods:The expression of IGF-I and IGF-IR in tumor and precancerous normal tissues were determined by tissue arrays and immuno- histochemical staining methods. Result :The expression rates of IGF-I ,IGF-IR proteins in colorectal carcinoma(64.58%, 58.33% )are significantly higher than those in precancerous normal tissues ( 16.67% ,22.9% ) ( P〈0.05 ). The over expression of IGF- I R was significantly related with the invasion degree, lymph node metastasis and Dukes classification in eolorectal cancer, but norelationship was found with other pathological factors ( P〉0.05 ). Conclusions: IGF- I R and IGF- I may he related closely with the development and progression of eoloreetal cancer. The up-regulation of IGF-IR may be related with Dukes stages and invasion degree of colorectal cancer. IGF-IR were very important for early diagnosis, evaluation of prognosis and target therapy of colorectal carcinoma.
