植物乳杆菌代谢产物和Ca^(2+)诱导人舌鳞癌CAL-27细胞凋亡的研究 被引量:2

摘要 目的:探讨植物乳酸杆菌发酵乳清蛋白获得的代谢产物以及Ca2+对人舌鳞癌CAL-27细胞生长抑制作用和诱导细胞凋亡作用。方法:植物乳杆菌23201经复原乳清菊糖培养基培养,制备相应的代谢产物1(LPM1)。利用草酸钠溶液沉淀LPM1中的Ca2+获得不含Ca2+的代谢产物(LPM1-F);采用MTT法检测不同浓度的LPM1和LPM1-F对CAL-27细胞体外增殖的抑制作用;通过吖啶橙染色法观察凋亡细胞的形态学变化;采用Annexinv-FITC/PI双染试剂盒检测早期凋亡率。结果:不同浓度LPM1和LPM1-F分别作用CAL-27细胞24h,最大抑制率分别达到(48.81±4.96)%和(56.70±2.33)%;荧光显微镜下观察,凋亡细胞皱缩,凝聚呈新月形或马蹄形,贴壁不良;流式细胞仪结果显示:经LPM1和LPM1-F处理的CAL-27细胞早期凋亡率均高于对照组。结论:植物乳酸杆菌代谢产物体外抑制CAL-27细胞生长,呈剂量的依赖性,并且诱导细胞凋亡,其中Ca2+对人舌鳞癌CAL-27细胞生长和凋亡无影响。
