莱菔硫烷对HCY致内皮细胞炎症损伤的治疗作用 被引量:4

摘要 目的:探讨莱菔硫烷(SFN)对同型半胱氨酸(HCY)诱导的人脐静脉细胞融合细胞EA.hy.926损伤的治疗作用。方法:将EA.hy.926细胞用10mmol/L的HCY诱导4h,再用0μmol/L~20μmol/L的SFN处理12h。采用噻唑蓝(MTT)法测吸光度,并计算粘附率;采用蛋白印记法(Western blotting)检测NF-κB的蛋白表达。结果:不同浓度的SFN均可抑制损伤后的EA.hy.926细胞的粘附和核转录因子κB(NF-κB)的蛋白表达量,并呈现剂量依赖关系。结论:SFN对HCY导致的EA.hy.926细胞炎症损伤有抑制作用,可能存在治疗作用。 Objective : To research the therapeutic effect of Sulforaphane of homocysteine - induced human um- bilical vein cells on the cell fusion EA. hy. 926 damage. Methods : The EA. hy. 926 cell 10mmol/ L HCY in- duced 4h, for 12h then 0μmol/L- 20μmol/L the SFN. MTT assay absorbance and calculate the adhesion rate; detect the protein expression of NF - kB by Western blotting. Results: Different concentrations of SFN injury EA. hy. 926 cell adhesion and nuclear transcription factor KB could inhibit protein expression, and a dose - dependent relationship. Conclusion: SFN can inhibit EA. hy. 926 of inflammatory injury caused by HCY and there may be a therapeutic effect.
