摘要
目的探讨夏枯草总三萜(TTP)对细菌脂多糖(LPS)诱导的RAW264.7细胞分泌炎症因子的调节及对Jak/Stat通路的影响。方法培养RAW264.7细胞系,体外用LPS(2μg/ml)刺激,ELISA法检测不同质量浓度的TTP(12.5、25、50、100、200μg/ml)分别作用24、48、72 h后对细胞分泌前列腺素E2(PGE2)的影响,选取质量浓度为25、50、100μg/ml的TTP,进一步用RT-PCR法检测肿瘤坏死因子α(TNF-α)、白介素6(IL-6)、Jak2和Stat3 mRNA的表达,ELISA法检测细胞上清中TNF-α、IL-6蛋白的表达。结果 TTP可以抑制细胞分泌PGE2,并且呈浓度和时间依赖性(P<0.05,P<0.01),TTP可以抑制LPS刺激的RAW264.7细胞分泌TNF-α、IL-6(P<0.01),并且明显抑制Jak2、Stat3的基因表达(P<0.01)。结论 TTP可以抑制RAW264.7细胞分泌炎症因子,调节细胞炎症因子网络的平衡,具有一定的抗炎作用,且这一作用的产生可能与Jak/Stat通路有关。
Objective To study the anti-inflammatory and immune effects of the total triterpenoids of Prunella vulgaris L. (TFP) in LPS treated RAW264.7 cell line and its possible mechanisms. Methods RAW264.7 cells were inhibited by TFP with different concentrations. After the cells were stimulated with LPS, cytokines protein and mRNA expressions were assayed by ELISA and RT-PCR. Results TIP effectively reduced the expression of TNF-α, IL-6 and PGE2 in LPS treated RAW264.7 cells ( P 〈 0. 01 ). It also inhibited mRNA expression of Jak2 and Stat3 (P 〈 0. 01 ). Conclusion TIP may participate in the process of anti-inflammatory response by inhibiting the ex- pression of some proinflammatory cytokines such as TNF-α, IL-6 and PGE2, probably through modulating the Jak2/ Star3 pathway.
