摘要 目的:研究G蛋白偶联受体(GPCRs)在人类肿瘤细胞中的表达,以及与人类恶性肿瘤发生发展的关系。方法:检索中国知识资源总库CNKI系列数据库、西文生物医学期刊数据库和PubMed数据库检索系统,分别以"G蛋白偶联受体(GPCRs)"和"肿瘤"、"生长抑素受体(SSTRs)"和"肿瘤"、"G蛋白偶联受体30(GPR30/GPER)"和"肿瘤"、"β肾上腺素受体(β-AR)"和"肿瘤"、"腺苷受体(AR)"和"肿瘤"、"胃泌素受体(CCK-BR)"和"肿瘤"为关键词,检索2000-01-2012-10国内外GPCRs与肿瘤方面的文献,CNKI中检索到21 207篇文献,西文生物医学期刊数据库中检索到8 317篇文献,PubMed中检索到5 674篇文献。纳入标准:1)GPCRs的信号传导通路;2)GPCRs与肿瘤的关系;3)GPCRs与肿瘤的靶向治疗。根据标准,纳入34篇文献进行分析。结果:GPCRs参与了胃癌、大肠癌、肺癌和乳腺癌等多种肿瘤的发生和发展过程,在肿瘤生长中起重要作用。GPCRs与人类肿瘤的增殖、侵袭和转移等多种恶性生物学行为密切相关,涉及到多种信号传导通路。GPCRs激动剂或者抑制剂与受体结合后,通过细胞内相应的信号传导通路能够影响肿瘤的生长,为临床肿瘤靶向分子治疗提供了提供实验理论依据。结论:GPCRs的研究能为人类肿瘤的诊治提供新的依据,也有利于发展新的抗癌药物作用靶点。 OBJECTIVE:To explore G protein-coupled receptors expression in human tumour ceil and its correlation with development of human carcinoma. METHODS: "GPRs" and "tumour","SSTRs" and "turnout", "GPR30/GPER" and "tumour","β-AR" and "tumour","AR" and "tumour", "CCK-BR" and "tumour" as key words were respectively searched by PubMed.CNKI and EMCC serises full-text database retrieval system from 2000-01 to 2012-10. There were 21 207 papers in CNKI,8 317 in EMCC and 5 674 in PubMed. The selection standards:1) The signal pathway of GPCRs; 2) The relationship of GPCRs and turnout; 3)GPCRs targeted therapy. According to the selection standrads 34 papers were analyzed finally. RESULTS: GPCRs participated in development of many kinds of tumors, such as Gastric cancer,Colorectal cancer, Lung cancer and breast cancer. GPCRs play an important role in tumor growth and associate with tumorigenesis,invasiveness and angiogenesis, and involve a variety of signaling pathways. Agonist or Inhibitor of GPCRs combined with receptors can affect tumor growth through intracellular corresponding signaling pathways, which provide theory basis for tumor targeted molecular therapy. CONCLUSION: GPCRs provide a new prognostic marker as well as a strategy of targeted therapy.
