重楼醇提物对小鼠免疫性肝损伤保护作用 被引量:6

Protective Effect of Ethanol Extract of Paris Ployphylla on Mice Model with Concanavalin A-Induced Acute Immunological Liver Injury

摘要 目的:观察重楼醇提物对刀豆蛋白A(Concanavalin A,ConA)介导的急性免疫性肝损伤小鼠的保护作用及其机制。方法:昆明种小鼠随机分为正常组、模型组、联苯双酯组(0.150 g.kg-1)、重楼醇提物低(1.3 g.kg-1)、中(2.6 g.kg-1)和高剂量组(3.9 g.kg-1)。采用ConA尾静脉注射法制作急性免疫性肝损伤模型。模型组、正常组给予生理盐水灌胃,其余组每日分别给予重楼醇提物、联苯双酯溶液灌胃,连续14 d。末次灌胃给药后1 h,除正常组外,其余各组按照15 mg.kg-1体重尾静脉注射ConA造模,造模给药后8 h处死动物取血或组织标本检测ALT、AST、SOD、MDA、GSH-Px含量;HE染色法观察肝组织病理学变化。结果:重楼醇提物能降低急性免疫性肝损伤小鼠ALT、AST和MDA含量(P<0.05或P<0.01);减轻肝组织病理损伤程度;升高SOD、GSH-Px活性(P<0.05或P<0.01)。结论:重楼醇提物对急性免疫性肝损伤小鼠具有一定的保护作用。 Objective : To observe the protective effect of ethanol extract of Paris Ployphylla on the mice model with Coneanavalin A-induced acute immunological liver injury. Methods: Mice were randomly divided into six groups:control group, model group, bifendate (0.15 g· kg-1 ), ethanol extract of Paris Ployphylla treatment low ( 1.3 g· kg-1), medium ( 2.6 g· kg-l ) and high ( 3.9 g · kg-1 ) groups. The acute immunological liver injury models were induced by injection of Concanavalin A from tail vein. The ALT, AST contents in serum, the activities of MDA, SOD and GSH-Px in the liver tissue were detected. Hepatic tissues stained with hematoxylin and eosin ( HE )were prepared for histological analysis on liver injury. Results : Ethanol extract of Paris Ployphylla could reduce the ALT, AST and MDA contents ( P〈0.05 or P〈0.01 ), and increase the SOD and GSH-Px activities in liver homogenate ( P〈0.05 or P〈0.01 ). Meanwhile, the pathological injury of hepatic tissues was alleviated. Conclusions: Ethanol extract of Paris Ployphylla showed significant protective effect on acute immunological liver injury in mice.
