摘要
目的探讨Rho激酶抑制剂法舒地尔对糖尿病肾病大鼠蛋白尿的影响。方法把30只SD雄性大鼠随机分为对照组(n=10)、糖尿病肾病组(n=10)、法舒地尔治疗组(n=10)。STZ腹腔注射造模成功后,法舒地尔治疗组给予法舒地尔10mg/(kg.d)腹腔注射,对照组及糖尿病肾病组给予等量的生理盐水腹腔注射,每天2次,连续12周。检测三组大鼠24h尿蛋白定量,取大鼠心脏血测定尿素氮(BUN)、肌酐(Scr)。结果糖尿病肾病组和法舒地尔治疗组24h尿蛋白水平明显高于对照组(P均<0.01)。与糖尿病肾病组比较,法舒地尔治疗组24h尿蛋白定量、血BUN、Scr明显下降(P<0.01)。结论法舒地尔能在一定程度上减少糖尿病肾病大鼠的蛋白尿、减轻肾功能的损害。
Objective To investigate the influence of fasudil on proteinuria in rats of diabetic nephropathy. Methods Thirty male Sprague-Dawley rats were randomly divided into normal group(n=10),diabetic nephropathy group(n=10) and fasudil-treated group(n = 10).Diabetic nephropathy was induced in male SD rats by STZ intraperitoneally injec- tion. After diabetic nephropathy (DN) model was established successfully. The rats were given fasudil 10 mg/(kg.d) in fasudil-treated group and the same amount of physiological saline in the other two groups once a day.After twelve weeks 24 h urine protein,blood urea nitrogen(BUN), serum creatinin(Scr) in three groups were detected. Results ①After administrating fasudil for 12 weeks in fasudil-treated group,the level of 24 h urine protein was significantly higher in diabetic nephropathy group and fasudil-treated group than that in normal group (P 〈 0.01 ). ②The level of 24 h urine protein, BUN, Scr were significantly lower in fasudil-treated group than that in diabetic nephropathy group (P 〈 0.01 ).Conclusion Fasudil can decrease the proteinuria in rats of diabetic nephropathy.
