秦皇岛海域产碱性蛋白酶菌株的筛选及产酶条件研究

Screening of strains producing alkaline protease from Qinhuangdao sea area and study of producing enzyme conditions

摘要 为了获得碱性蛋白酶产生菌株并优化其发酵条件,试验采用初筛和复筛方法从秦皇岛鸽子窝采集的泥样中筛选产碱性蛋白酶菌株,同时考察pH值、培养温度、培养时间、装液量等单因素对菌株产酶能力的影响,进而进行4因素3水平的正交试验分析,最后考察甘油、蔗糖、乳糖、葡萄糖和淀粉对菌株产酶的影响。结果表明:得到1株产碱性蛋白酶能力较高的菌株SD13,其较佳发酵条件为pH值为9.0,温度为37℃,培养时间为48 h,装液量为25 mL培养基/50 mL三角瓶,且影响的主次顺序为pH值、温度、培养时间和装液量;最适碳源为葡萄糖,最适添加量为1%;菌株SD13在较佳条件下发酵液的最大酶活力达189.912 U/mL。说明可以从秦皇岛海域获得产碱性蛋白酶菌株。 To obtain strains producing alkaline protease and optimize the fermentation conditions, screening and rescreening methods were used to screen stranis producing alkaline protease "from sea muds samples collected through Qinhuangdao pigeon nest park. Single factor experiments, such as pH, culture temperature, culture time, and the volume of liquid were performed to investigate the effects of these factors on enzyme pro- duction capacity of the strains. Then the orthogonal array for four factors and three levels was used. At last, the effects of glycerin, sucrose, lac- tose, glucose, and starch on the enzyme ability were investigated. A strain SD13 having high ability to produce alkaline protease was obtained, and its optimal fermentation conditions were p.H value of 9.0, culture temperature of 37 ~C, culture time of 48 h, and liquid volume of 25 mL medium/50 mL Erlenmeyer flask. The primary and secondary factors affecting the" enzyme production were as followed : pH, culture tempera- ture, culture time, the volume of liquid. The optimal carbon source was glucose, and the optimal addition of glucose was 1%. The maximum en- zyme activity of fermentation broth for the strains SD13 reached to 189. 912 U/mL under the optimal conditions. The results showed that the strains producing alkaline protease could be obtained from Qinhuangdao sea area.
