大鼠可逆性局灶性脑缺血模型复制方法的改进 被引量:13

摘要 目的 寻找操作简便、创伤较小的建立可逆性局灶性脑缺血模型的方法。方法 在现有线栓法的基础上作如下改进 :大鼠体重与线栓直径的匹配以及线栓插入深度的控制 ;线栓前段覆左旋多聚赖氨酸和肝素 ;麻醉后拔线。取脑组织作TTC染色和 HE染色 ,并进行神经功能缺陷体征评分。结果 左侧大脑中动脉缺血区经 TTC染色颜色苍白 ,经 HE染色呈缺血性改变。不予再灌注其评分进行性增加 ,给予再灌注则评分呈不同程度下降。结论 改进后的线栓法复制的可逆性局灶性脑缺血模型是研究脑缺血再灌注损伤较为理想的模型。 Objective To search for a simple and less invasive method for developing a reversible focal cerebral ischemia.Methods On the basis of present thread embolism method,the following modification was made:matching the weight of rats and the diameter of the thread,controlling the insertion depth;coating the thread with Poly L lysine and heparin,pulling out the thread after anesthesia.Brain tissue was taken out to undergo TTC and HE staining and neurological deficit sign scores were performed.Results The brain tissue supplied by left middle cerebral artery was pale under TTC staining and showed histologically ischemic changes under HE staining.The neurological deficit sign score under no reperfusion was increased progressively,while the score under reperfusion was decreased to some extent.Conclusion The reversible focal cerebral ischemia model developed by the modified thread embolism method is an ideal one to study the injury of cerebral ischemia/reperfusion.
