摘要
目的:观察加味丹栀逍遥散对抑郁模型大鼠血浆内CORT和ACTH含量的变化,探讨加味丹栀逍遥散对抑郁模型大鼠下丘脑-垂体-肾上腺(HPA)轴功能的影响。方法:将50只Wistar雄性大鼠随机分为空白对照组、模型组、氟西汀组、加味丹栀逍遥散低、高剂量组,每组10只;采用孤养结合慢性轻度不可预见性应激的复合模型,除空白组外,其余各组均接受不同应激因子刺激21 d,建立慢性应激抑郁大鼠模型,随后分别按氟西汀组2.5 mg.kg-1、加味丹栀逍遥散低、高剂量组2.04,4.08 g.kg-1,各组均按10 mL.kg-1ig给药,空白对照组和模型组ig 10 mL.kg-1生理盐水1次/d,每天应激前灌胃,从造模第22天开始,至动物处死当天,共21 d,实验周期共42 d;采用放射免疫分析法(RIA)测定大鼠血浆皮质醇(CORT)和促肾上腺皮质激素(ACTH)的变化。结果:,与空白组比较,模型组大鼠血浆CORT(33.22±3.84)ng.L-1和ACTH(203.78±12.69)ng.L-1的水平显著高于空白对照组(P﹤0.05),说明抑郁模型动物存在HPA轴的异常兴奋;经过治疗后,各组大鼠CORT和ACTH的水平明显低于模型组水平(P﹤0.05)。结论:加味丹栀逍遥散可以通过降低血浆中CORT和ACTH的水平,拮抗应激导致的HPA轴亢进从而起到治疗作用。
Objective: To investigate the effects of Jiawei Danzhi Xiaoyao powder on the concentraion changes of adrenocorticotropic hormone (ACTH) and corticosterone (CORT) in blood plasma of depression model in rats, and discuss the effect of Jiawei Danzhi Xiaoyao powder on the function of HPA in the depression rats. Method: Depression model of rat was produced by using chronic unpredictable mild stress stimulation combined with lonely raising for 42 days.The first 21 days is built model,From the twenty-second days of models,the rats received the stress and medicine intervention.Use radioimmunoassay(RIA) to determine the change of cortisol(CORT) and adrenocorticotropin(ACTH). Result: The concentraions of CORT and ACTH were increased significantly in blood plasma of the model rats compared to that of the normal group(P〈0.05);also the concentraions of CORT and ACTH were reduced significantly in blood plasma of Jiawei Danzhi Xiaoyao powder group compared to that of the model group (P〈0.05). Conclusion: Jiawei Danzhi Xiaoyao powder can reduce the concentraion changes of CORT and ACTH in blood plasma of depression model in rats, thereby play its treating role.
