摘要
目的观察红花注射液(ICT)对2,4,6-三硝基苯磺酸(2,4,6-TNBS)所致大鼠溃疡性结肠炎(UC)的疗效及其对结肠组织中EGF和TGF-β1表达的影响。方法将大鼠随机分为3组:模型组、ICT组和正常对照组(n均为10)。用TNBS灌肠复制大鼠UC模型,用ICT干预治疗,观察大鼠疾病活动指数(DAI)及大肠组织学损伤以判断疗效;用免疫组化法检测UC大鼠结肠黏膜EGF和TGF-β1的表达,用逆转录-聚合酶链反应(RT-PCR)法检测EGF mRNA和TGF-β1mRNA的表达,从而探讨其治疗机制。结果与模型组相比,ICT组的DAI及组织学损伤显著改善(P<0.05);与正常对照组相比,ICT组与模型组大鼠结肠组织中EGF,TGF-β1蛋白及其mRNA的表达均显著增高(P<0.05);而与模型组相比,ICT组结肠组织中EGF及其mRNA的表达均显著增高(P<0.05);TGF-β1蛋白及其mRNA的表达显著降低(P<0.05)。结论 ICT对TNBS所致大鼠UC具有一定疗效,ICT可以增加EGF,TGF-β1的表达,促进创伤组织的修复。
Objective To investigate the therapeutic effects nf injection of carthamus tinctorius(1CT) on ulcerative colitis(UC) induced by 2,4,6-trinitro -ben-zene-sulfonic acid(TNBS) in rats as well as its effect on the expression of epidermal growth factor(EGF) and trans forming growth factor-β1 ( TGF-β1). Methods Thirty male Wistar rats were randomly divided into treatment group,TNBS control group and normal control group(n = l0 ). The rats in treatment group were treated with injection of earlbamns tinctorius(ICT). Curative effect was judged by investigate the disease activity index( DAI) and histological damage. The expression of EGF and TGF-β1 in cohm tissue was detec ted hy immunohistochemistry(IHC). And the expression of EGFmRNA and TGF-β1 mRNA were detected by reverse trauscription-polymerase chain reaction( RT-PCR ) method. Results Compared with model group,the DAI and histological danmge of colon tissues in treatment group were significantly improved. The level of EGF and TGF-β1 in model group arid treatment group were significantly higher than that in normal control group ( P 〈 0.05 ). The level of EGF and TGF-β1 in treatment group was significantly higher than that in model group ( P 〈 0.05 ). The level of TGF-β1 in treatment group was lower than that in model group( P 〈 0.05 ). Conclusion This stt, dy indicates that the ICT can relieve colitis in rats induced by TNBS and inc.reasing EGF and decreasing TGF-β1 may be one nf its functional mechanism.
