摘要
本实验用Balb/c小鼠、腹水型肝癌(Hca-F_(25))尾静脉接种所产生的肺转移瘤和淋巴结转移瘤分别重复循环筛选10次(即肺瘤结→尾静脉→肺瘤结……;和淋巴结瘤结→尾静脉→淋巴结瘤结……)得到同一肿瘤内具有不同转移倾向的两个瘤细胞亚群P_(10)(肺转移瘤第10代)和L_(10)(淋巴结转移瘤第10代)。随着循环筛选代数增加,肺转移亚群的肺转移率显著升高,淋巴结转移亚群也有相似变化。经尾静脉,腋皮下和脚掌三途径接种,在前两途径接种中,P_(10)的肺转移率均显著高于L_(10);L_(10)的淋巴结转移率显著高于P_(10)。脚掌接种,P_(10)和L_(10)的转移率无显著差别。两亚群的肺转移灶大多形成于小血管或小支气管周围。细胞侵袭性试验表明P_(10)和L_(10)各自对肺和淋巴结粘附,侵袭增强,可很快侵入深部组织生长。本实验结果提示肿瘤转移的器官亲和性不仅与瘤细胞本身生物学性状有关,也与肿瘤移植部位有关。
After the suspension of ascitic Hepatoma (H_(22)-F_(25)) was injected intotail veins of Balb/c mice, the metastases in lungs and lymph-nodeswere collected separately. Following courses were repeated 10 times:lung metastases→tail veins→lung metastases……; lymph-nodes metas-tases→tail veins→lymph→nodes……. Finally, two substrains with dif-ferent metastatic tendencies in same tumor were gotten (P10, the tengenerations of lung metastatic substrain; L_(10), the ten generations oflymph-nodes metastatic substrain). Compare the metastatic percentageof parent strain with 7 th generation and 10 th generation, we foundthat with the increase of selecting times, the metastatic rate of lungsubstrain in lungs increased gradually. Similar manifestation occurredin lymph-node substrain. Among three injection ways, (tail vein, subcutaneous, foot pad), except the later way, the metastatic rate ofP_(10) in lung was higher than that of L_(10) obviously; and the metastaticrate of L_(10) in lymph-nodes was higher than that of P_(10) obviously.The anatomic distribution of clones in lungs were similar between thetwo substrains, they mostly grow around minute blood vessels andbronchia. The test of cell invasion suggested that P_(10) and L_(10) showedstronger adhesive ability and invasion ability to lungs and to lymph-nodes respectively. There were different ultrastructural characteristicsbetween P_(10) and L_(10). This article suggested that the organ affinityof tumor metastasis associated with not only biological characteristicsof tumor cells, but also with the location of tumor transplantation,invasion ability to target organs and adaptation of growth.
